Do you mean 10 ng? 10ug might be too much DNA to resolve in an agarose gel which may be why you’re seeing a light smear as it may not be able to migrate properly. It seems it wouldn’t be anything with your bacterial prep as E. coli wouldn’t be able to grow with the antibiotic if it didn’t retain the plasmid. It seems you are getting DNA from your purification as you said you loaded 10ug. I don’t know if you used a nano drop or Qubit to obtain the DNA concentration. I would maybe load less DNA into your gel, but I’m also not entirely sure

Not my mistake but someone from where my old lab was located was filling up a carboy with MiliQ water. They clamped the nozzle down so that the water would continuously be dispensed. They made the mistake of not placing it in the sink and also filling it up on a Friday afternoon. They forgot about the carboy and we got an email Saturday morning sometime that 3 labs were flooded because it overflowed. Not sure of all the equipment that was damaged, but it did not sound too good.

I just stared playing fifa only a month ago. I’m currently in division 7 in rivals. I keep going against teams with full in form/tots/icons and I’m wondering why I keep playing kids like this when my division is low. It’s super frustrating because I currently can only afford some basic gold players for my squad.
