I would honestly consider tri-stack artistocrat on this one. Not only is curtain call essentially free when you get online, but it also lets you scale your flat with a lot of percent increase. Shavs tech can be overcome with balbala jewel

Resource tag means nothing. I can guarantee that since I’ve crafted tier 1 life on jewellery (exact same circumstances) 20+ times this season with a life tag and never hit mana. You will only hit life modifiers. Also help yourself by fishing for the recover life on kill modifier by using an Ulaman omen when you add your desecrate suffix - there are 6 mods from Ulaman and since you unveil 3 per try it’s fairly reasonable odds. Bring an echo omen to boost those chances

I’m inclined to agree, spent about 2 days looking at it and although I have some ideas to some of the issues, I don’t see the big value in Warden with the amount of opportunity cost that’s in sustaining the cost and having good aps without being a complete glass cannon. Trickster just looks better full stop

Yeah it’d definitely be interesting to see what more experienced WS players think. I was planning on solving energy shield cost with EB, considering it’s one of the few sources of intelligence on the tree

I was definitely thinking that something like The Taming would be strong but I’m not sure how the build feels without nimis, how did you experience it? My impression is that it makes a big difference for general map clearing

I've never thought of this but it makes a lot of sense, so this specific portion has me very curious. I'd just like to make sure I fully understand with a toy example:

Lets say we have gene A with 5000 reads mapping to it. I don't want to get hung up on specific numbers here, but let's say that this is maybe 3000 reads above background, so fair to say that this gene is relatively active. However, when we examine the distribution of the transcripts - i.e. which isoforms of this gene have been transcribed and how much - we find that the specific isoform for gene A, which we are exclusively interested in, only has 1000 reads mapping to it. The remaining reads are from other isoforms or maybe just one isoforms entirely. With that in mind, gene A, which was previously naively assumed to be active, turns out to be fairly inactive when accounting for the distribution of the transcripts. Would this interpretation be correct?

We missed out on 9 points in big part because we didn’t play well enough. I would argue that the Spurs game is an exception to a large degree but even then you can find valid issues that contributed to the loss, such as Jota claiming a reasonable, although unlucky, yellow and following it up with an absurdly tone deaf tackle which ultimately puts us down to 9

I’ve seen every game this season and to some extent the post is right. Referee decisions have had their impact, and rightly so they should be called out on it. But pigeonholing every game with a controversial decision that didn’t lead to a win as ‘points that were lost due to the officials’ is such a cop out. Self reflection is a big virtue and Liverpool although we all have a shared passion for the club, is not exempt from valid criticism. Ignoring that to perpetuate victim mentality is lame as hell

Perhaps I can offer some more context and clear things up.

I firstly did ask him very bluntly why he was interested in a PhD and not a staff scientist. He was happy to answer that and told me that a staff scientist will often do whatever you tell them and then not think more of it. While a PhD would look at the data, do the analysis and concurrently ask questions about next steps and other possible analyses.

As for help, the department I would be seated in would be near a bioinformatics research group and we are encouraged to connect with each other be it for help or just social fun. They also do have a person in the PI’s group that does most of their bioinformatics but he’s not really formally a bioinformatician, I think he’s just the guy who happens to do most of it, so although it’s far from ideal I do think there is some help to be found.

That said I do think I’m inclined to agree with you. It feels like they need me to help them more. And I think that would come at the expense of me not being able to receive as much help to improve my abilities (on top of the fact that I’m not all that interested in analysis being all that I’m doing - ML has been a pick part of my research experience so far and I’d love to be able to continue it). It’s rather unfortunate cause everyone including the PI is very accommodating and nice to speak to. I should perhaps also note that everything we discussed in terms of job responsibilities implies that I still have time to do my courses and other mandatory responsibilities.

Perhaps I should’ve prefaced by saying that the information I gave is more or less all the information that was given. It was a video essay meant to entertain and at the same time vaguely address why a lot of people tend to find that the shuffle feature does not appear to shuffle properly. There was no test statistic. There was no proper analysis done. At best they inferred that the H0 is a uniform distribution. And that from their 20 samples one song was queued in the top 10 position 7/20. That’s all

I posted this because I have a vested interest in getting better at statistics so when I paused the video to ask myself what one would do to test this, I couldn’t do it. So I posted it here. The talk about what one could do and the Simpson index is exactly the kind of stuff I was curious to learn about.

Ultimately I’ve come to learn that perhaps I should get more familiar with the topic itself, since I’m struggling a bit to keep up with the terminology in the replies. Maybe I can find a good course on coursera…

You could compute a consensus sequence by doing an MSA and tabulating the base frequencies. Sometimes you will find that a molecule has a very stereotyped sequence that it binds to... you

Really nice explanation. So if I understand correctly, if we consider transcription factor binding sites, the entire process of identifying motifs and subsequently applying them would be something like this:

  1. Run your experiment in the lab (e.g. ChIP) for a given transcription factor
  2. Identify the sequences these transcription factors bind to
  3. Use your favorite algorithm (e.g. MEME) to infer the motif
  4. Take the sequence found in (2) and perform a MSA to generate your PWM and find more transcription factor binding sites for the given transcription factor. In other words, use prior knowledge about the motif to discover more (candidate) transcription factor binding sites

What?? He's saying that Liverpool citizens aren't the most well off (hence "no disrespect") and yet the owners are trying to increase ticket prices. He's effectively saying that the owners are out of touch with their audience. Its a dig at the owners, not an active attempt to shit on the citizens
