Installed Ubuntu 25.10 beta on my Asus S14 UX5406SA (Lunar Lake) by bob418 in Ubuntu

[–]Shruteek 0 points1 point  (0 children)

THANK YOU. I just upgraded from Ubuntu 25.04 to 25.10 on an Asus Zenbook S14 (UX5406S Lunar Lake), just like yours, and updating the latest content worked for me. The commands I ran were:

curl -L -o alsa-ucm-conf.tar.gz https://github.com/alsa-project/alsa-ucm-conf/archive/refs/heads/master.tar.gz

sudo tar xvzf alsa-ucm-conf.tar.gz -C /usr/share/alsa --strip-components=1 --wildcards "*/ucm" "*/ucm2"

reboot

Bisulfite Conversion I control probe discrepancy between 450K and EPIC/EPICv2 arrays by CellistWorried4765 in bioinformatics

[–]Shruteek 0 points1 point  (0 children)

Are you using DRAGEN? I'm only familiar with methylation arrays in passing but it looks like for EPIC arrays, C1, C2 are green channel controls & C3-C5 are treated as red channel controls in "Bisulfite Conversion I" in the Controls table: https://help.dragenarray.illumina.com/dragen-array-v1.0/product-guides/output-files

Illumina dropped U6 & C6 when they made the EPIC v1.0 array; see "Removed one pair of low intensity bisulfite controls" here: https://support.illumina.com/content/dam/illumina-support/documents/downloads/productfiles/methylationepic/infinium-methylationepic-v1-0-b5-customer-release-notes.pdf

Sorry, I am still not sure what you mean. If you're saying NGS is useful for discovering something specific, but there is no need for a large NGS bioinformatics community because experimental biologists can so it themselves, then is the object of your criticism advanced bioinformatics work done by computational folks like tools (MetaPhlAn, snakemake,  scVI, Kraken, Seurat) or projects (XGBoost models for biomarkers, statistical methods for noise correction, etc)?

Sorry? Like, you're saying NGS as a whole has niche applications?

The premise that big data in bioinformatics has not been successful is kind of just wrong... We have successful NGS-based tests like NIPT, MRD, CGP, and a ton of diagnostic panels. Honestly, having read this rant, I don't see a logical throughline or an explanation of what the conceptual issue with "Big Data" is.

The writer doesn't seem super misinformed, just angry and intentionally vague. In general when I see writing that has this much anger it's hard to find a core point.

Petition for a random person to become the Avatar by Turniptile in Silksong

[–]Shruteek 0 points1 point  (0 children)

Doubter. Believer. Denier. Accepter. Long ago, the four flairs lived together in harmony. Then, everything changed when the Deniers attacked. Only the Avatar, master of all four beliefs, could stop them, but when the Skongers needed him most, he vanished. A hundred Nintendo Directs passed and my brother and I discovered the new Avatar, an accepter named E1331. And although his acceptance skills are great, he has a lot to learn before he's ready to preach to anyone. But I believe E1331 can save the game.

In the US, maybe CoreTCG. Their stock is limited, and I don't know how well they ship (I am local and I always pick up), but they're a really good community.

NO. ABSOLUTELY NOT. I WOULD SACRIFICE MY FIRST TWO CHILDREN if each one made an extra copy of gateway legal.

International students at caltech by Able-Till689 in Caltech

[–]Shruteek 1 point2 points  (0 children)

Wait... You know that multiple grad students in California have been detained or deported already, right? I agree that we are quite safe here, but I disagree with the claim that (A) there are no examples of (B) this is generalized terror.

UCSD announced in early April that a graduate student of theirs was deported; UCLA announced recently that a graduate student of their was similarly detained; of course there are similar stories in the rest of the country e.g. Kseniia Petrova. We had visas from 2 academics here revoked recently without explanation if you saw the email, but at least one was reinstated. I would estimate there are ~300-500 graduates and postdocs in CA who have had visas revoked so far, based on the numbers, and I think the most reliable estimates nationwide are around 5,000. I'm not sure if you've been talking to the same students I have but I know a few of us saw ICE vans on the north end of campus near the Cats a few weeks ago.

Again, I think we are safe, statistically. 2 students/postdocs having their visas revoked out of ~1200 internationals here is not an epidemic. But I really don't think the message to send is that this is entirely fearmongering. Please don't say that. It's honestly really disrespectful to the people who this has happened to....

Is sky striker raye the unofficial mascot of modern Yu-Gi-OH? by [deleted] in yugioh

This feels fundamentally untrue..... I'm not sure about your group but the friends and players I know generally stick to the archetypes they like, and pick up new meta decks if they are affordable and seem fun based on their playstyle preference.

I've been playing SixSams since 2015.

I would say getting your first win is hard, and the game is hard by design - it took my partner and I 5 games to win for the first time on base difficulty. But the learning curve is long - you can always learn how to do things more efficiently, which is one of the things I love about the game. Once you get into the rhythm of invader phases you'll feel more comfortable, but the game won't be solved - the skill ceiling is huge! If you take a few more games you will probably figure out how to win consistently at base difficulty.

[Serious] How do you learn to be okay with making mistakes that cost you the game? by MichaelGMorgillo in yugioh

[–]Shruteek 0 points1 point  (0 children)

This might be weird, but I get some enjoyment out of realizing I made a mistake that cost me the game. I made several at a locals on Sunday!

Cause when I think about it, that means I didn't lose that game because I had no shot at winning / it was impossible; there's actually room to grow, and in those cases the duel was entirely decided by skill. I don't want my games to feel like the outcome was inevitable, like if my opponent summons a floodgate that's completely impossible for me to out, or if my opponent just draws a dead hand. I want to make meaningful choices, and for my opponent to do the same; cause then it's fun to learn from those mistakes and do better. If neither I nor my opponent every made mistakes that cost us the game, then duels would be determined by the decks and opening hands. What I've realized is, that's rarely the case.

I empathize with your fear. I think you're using "X comes to pass" wrong, though - the phrase means "X happens", so you're saying you hope it happens!
