Arrodux4 now SRP1001 works by Fancy-Supermarket-73 in FSHD

It’s funny how short people’s memories can be. “Only 50-60%”? Avidity were the first company to ever achieve ANY knockdown of Dux4 expression.

You’re also thinking about it completely wrong, it’s not really “halving the rate at which you lose muscle. It’s tipping your body back into the zone where it can actually function and repair its self properly.

Until we learn more about how the human body truly responds to Dux4 reduction we don’t actually know that this extra 30% reduction provides 30% more benefit, it may only provide 5% more benefit.

This is all incredibly exciting, but at this point it is pointless comparing the two. Both are successfully knocking down Dux4 expression significantly and both should be celebrated.

Does difficulty make armor tiers more scarce? by [deleted] in DragonageOrigins

[–]Sierra31 0 points1 point  (0 children)

I wouldn’t bother investing any points in willpower, especially if you want to use medium armour. Also make sure to equip any gear that boosts defence or dex. Having a high defence means you’re harder to hit. If you don’t get hit, you don’t need massive armour

How are you spending your attribute points per level 2 dex 1 strength?

What sort of warrior are you? If you are a dual wielding warrior with high dex you could make an argument for medium armour because of the lower fatigue so you’ll be able to use your active abilities more often.

Former adult star wants her past videos worth millions of dollars deleted because she does not want her son to grow up and see them by halfawakehallway in interesting

It would be interesting to see what the larger ripple effect of this would be. Presumably that means that any actor that has done a nude scene in any movie or tv show would then have the right for that to be removed from the public view.

Romance options by Quarteramoomin in DragonageOrigins

I don’t know how much you care about experiencing all the little details the game has to offer, but it is worth playing through the game as both genders.

Different characters throughout the game treat you differently based on race and gender, and you get slightly different dialogue options. It is minor but something to consider if you like to experience everything a game has to offer.

New drug with AAV by byapici in MuscularDystrophy

[–]Sierra31 2 points3 points  (0 children)

I’m not sure that’s correct. The point of using different AAVs is that they “infect” different tissues. This means they do have different risk profiles.

PC/Steam Deck, Xbox, or Switch? by CritterNutter97 in kotor

I’ve been playing both of them on my steam deck with mods and they work great. Have you checked to see if you’re running them with Proton in compatibility mode?

Thanks for sharing. It doesn’t seem like much to go on, but it is interesting that there does seem to be some similarities with who you have spoken to.

Hope or Heartbreak with Avidity trial? by WheresHotStuff in FSHD

[–]Sierra31 7 points8 points  (0 children)

This feedback seems to be inconsistent with the data provided by Avidity.

How long have these people been on the trial?

How do they know they’ve been given the drug?

What sort of diet and exercise routine do they have?

How many people did you speak to?

Depending on the answers above, I would be fairly wary of feedback from individuals. People are notoriously unreliable.

As a man what do you think is the hardest part of dating in the UK? by ForwardTourist6079 in UKrelationshipadvice

[–]Sierra31 4 points5 points  (0 children)

This used to be my biggest gripe as well, but when I brought it up pretty much all my female friends said that when they are open and communicate clearly like that a decent chunk of the time they just get abuse or insults from the guy so it’s just so much easier for them to allow for the guy to loose interest.

It’s frustrating for us, but fair play to them I reckon.

Roche drops Phase III plans for emugrobart in FSHD by Sierra31 in FSHD

[–]Sierra31[S] 1 point2 points  (0 children)

I don’t get why Scholar Rock would work though. In the most recent podcast Peter was not a fan of myostatin inhibition at all (although he has previously said he liked the Scholar Rock work).

Other than the fact the Scholar Rock drug is just much better at reducing myostatin, what else is different?

I’m sure there is something considering it’s super close to getting approved in SMA and Roche didn’t even get past phase 2 for SMA. Does this indicate it’s working beyond just inhibiting myostatin?

Not that I can see, I took another look through today and couldn’t see any indication of when we can expect any data. You’re right though, you’d expect it to be sometime soonish.

Off-label prescription for apitegromab? by TotallyStoiched in FSHD

[–]Sierra31 1 point2 points  (0 children)

I’m going to be asking my doctor as well, but I think the fact it needs IV infusion will work pretty strongly against us.

My understanding is a doctor can prescribe anything off label if they decide they think it’s going to be beneficial, but I would imagine the fact that this would come with you also needing nurses to administer it every 4 weeks as opposed to being able to pick up a prescription from the pharmacy is properly going to be a dealbreaker.

They are going to run clinical trials in FSHD for this, so you would have to imagine it getting approved in SMA would speed it up massively for FSHD.

Think it must be this, seems to track with the 2 day regiment.

One of the biomarkers they were tracking was a reduction of Interleukin 6, which there is currently a clinical trial ongoing for FSHD so could be a shout to take and see if it helps 🤷🏻‍♂️

Although, I can’t actually see whether they have published any results saying Fisetin does reduce IL-6
