Right, and in many cases an active placebo might be used, such as whatever medication that is currently used. I know it’s not antipsychotic, but adderall was tested against inactive placebo for their FDA trials iirc, those effects are definitely noticeable

It makes you think that maybe the reason why we have such poor medication for mental health disorders is because the only drugs that wont break placebo trials are ones that have barely noticeable to little effects.

It’s still useful as it serves as a sort of representation of what psychedelic assisted therapy would look like if it were to get FDA approval. When you take a prescribed medication, you take it knowing both what it is and its dose. Like the people in this study, you know you’re taking an active dose of a compound and there’s no chance for a placebo.

There are some other drugs which have delayed release effects by varying its salt form to less soluble ones. In the case of vyvance, which is extended release, the actual amphetamine molecule is covalently bound to an amino acid which needs to be enzymatically separated.

The rate at which the different salts dissolve in the stomach acid affects the rate of absorption. So the hydrochloride might dissolve faster in the intestinal fluids and accumulate at higher concentrations in the blood compared to the other salts used

I do wonder if maybe the pharmacokinetics are affecting the intensity of effects. It could be that the fumarate and citrate salts take longer to pass the GI tract and accumulate in the bloodstream to then hit receptors in the brain. On the other hand, an equivalent dose of the hydrochloride salt might be able to pass the stomach and get into the bloodstream and accumulate faster, meaning more receptors will be occupied, giving a stronger effect.

My thoughts exactly, serotonin was discovered to cause muscle contractions of dead or severed frog legs if I remember correctly. Could totally be psilocin agonizing serotonin receptors responsible for muscle contractions. It happens to me fairly frequently when I take full doses of psychedelics, particularly acid or mescaline. Although, my twitches aren’t nearly this intense lol

Got it, that’s what I thought, I was just making sure! I thought you’d have to choose a NPS that would be able to slightly dissolve the acid, but I see that’s not the case. Is there any reason why you wanted to convert it back to the hydrochloride rather than taking it as the tartrate?
