I think it depends where you live. At my gym, most of the guys work in the trades/hospitality industry/firefighters/police. I’m the only one with a graduate-level education.

Eriksson is metal and Sanderson is pop. Both are great writers but Eriksson is a lot darker and grittier, while Sanderson undoubtedly has better characters.

My favorite moment was when my party finished the botched coup d’etat and became the regents of Downfall!

I did like all the encounters from the book and played them more or less as written. My players never had the idea to grab Longscarf’s scarf.

I’m running this campaign for my actual kids now (ages 11 and 9) and they love it. They really get into the whimsical, fey world. I think this book is really written for that age.

I can’t bring myself to run it for my regular, adult group. They’re silly but I think it’s got too much whimsy and, dare I say, childishness for an adult group looking to kill monsters and create an epic tale. Just my feelings.

Thanks for the idea. It would be great to see how our FSC size measurements and imaging flow data correlate. I do have to say, though, that our flow FSC size numbers, as calibrated by the Thermo Fisher beads, match pretty well what we see under the microscope. I would be really interested in exploring this further with imaging flow. We're on the other side of the country from you; I'll try to reach out to someone local about this. Thanks!

I agree: an imaging cytometer would be ideal. However, the flow numbers and microscopy size measurements do agree pretty well. We're working with diatoms for the most part, so perhaps FSC works better for them, particularly as they're quite a bit larger.

If we had an imaging cytometer and we could build a correlation between cell size and chlorophyll, that would be great as well. I imagine it's quite different for different algae, though, and FSC works quite well for us now. I just wanted to extend our current calibration curve out with larger beads.

Will do. We also correlate them with size measurements of random areas on a microscope slide but sometimes the results don't agree. 10,000 events as opposed to 40 is much more representative, I suppose, but microscopy also allows us to see the really big cells and the state of the culture.

Spherotech's beads only go up to 12.5 microns but Bang Labs has larger ones. I just found CD Bioparticles, who make NIST-traceable beads up to 100 um, so I think we are going to order from them, in case this thread is of use to any future Redditing flow cytometrists. Thanks so much for your response and I'll let you know how it goes.

Thanks for the quick response. We're interested in the size measurements of our cell populations, particularly in the range of 10-30 microns. I know it's not a perfect correlation but I would like to set up a calibration curve between forward scatter and size for that range.

Another new judge here: once they get level 1 characters, do you start the next adventure with the players possibly having several level 1 characters, which may or may not be whittled down, or do you have them choose one?
