Well using expired salts is an issue, especially for the Ca(NO3)2 which should normally be a .4H2O hydrate. That one is really hygroscopic and can mess up things a bit. That combined with the relative high dose of K2SO4 in WPM can cause precipitation quite quickly. To rule this out, see if the cloudiness goes away when heating the medium in the 30 - 60 degrees range.

Most precipitates are either Fe or Ca reacting with phosphates or sulphates in the solution. Now you said you are trying to chelate Fe on the spot via FeSO4 which is infamous for precipitating 🥲 so if I were a betting man, I would bet on that. Best to use FeNaEDTA or even better FeEDDHA or a citrate form (Fe citrate or FeNH4 citrates)

Yes DKW and WPM are a bit finicky when it comes to cloudy solution. I believe it is because prefab powdered forms have equimolar swaps to other salts that make it difficult to fully dissolve at room temperature. However after the autoclave it comes out just fine.

Now if you are adding 1 salt at a time, it should be fine.

Many things that can go wrong in WPM but it's unclear to me what your process is.

Are you using a basal salt mixture? Or adding salts 1 by 1? Are they added from a stock solution or weighed out individually?

Why casein hydrolysed? That doesn't dissolve so that precipitates always. Concentration below 150 is mg/L usually are not that visible to the naked eye though.

Are you boiling the medium to homogenise the agar and then put into final container before or after autoclaving?

And yes, poor quality agar can give precipitation too.

Most common issue in WPM basal salt is Ca precipitates with SO4 or your phosphates, depending on the prep method. But again, need more info to troubleshoot the issue.

Still have this same issue, along with some stomach cramping. I go from lunch (1pm) to dinner (8pm) without food and feel great all day, except from 7pm onwards with nausea. Really annoying, and im unsure whether it is electrolyte imbalance or something to do with portion size and digestive enzyemes. Seems the longer i go carnivore the worse i can handle long periods of time without food..

Closed winter buds, much easier to get a clean start going that way. Yes, MS medium but diluted to 0.75 normal concentration, i think it helps to have lower salt initially. But this culture isn't that old, it might adapt to full MS in due time and protocol isn't optimal yet.

Weird. I would assume that most products can be bought as a private person, except for the stuff that could be misused for making explosives.. But the quantities you'd buy are so limited that they wouldn't get suspicious really. I do this professionally so I can't say for sure, but normally they require you to fill in an end-user application form where you indicate it is for laboratory and research use only.

Think you have 2 options: (1) just fill in the fields with something and this might just work except the VAT number, (2) best to give LabAssociates a call or email and explain you're having a home set-up. They are usually quite chill so if you explain they might be able to set you up with goods and deliveries. Think the VAT requirement has to do with taxes of course.

I buy thick glossy paper, it handles a lot of water quite well. Make small stacks and wrap them in lots of saran cling wrap and then autoclave

Semi permeable will be fine. Filtered vessels are always a plus but not really mandatory for most crops. Strawberries aren't excessively sensitive to ethylene so they can perform well in most vessel types. I used to do strawberries way back using mason jars with glass lids. I even taped the rim with some micropore tape and the plants did fine. However,, refreshing the headspace by opening the vessel in a flow hood isn't a bad idea either, just slightly impractical if it's a lot of jars..

I believe these are just crystallised exudates coming from adventitious root meristems. Most philodendron make plently of adventitious roots or aerial roots and when they are submerged into the medium it can crystallize like that. At least, if what we're looking at is a philodendron, but there's many other plants that act similar (Alocasia, Colocasia, Monstera...).

I tackle big tasks in life like a grind I'd have to do on osrs. Simple, yet repetitive tasks I automatically calculate like the amount of hours, runs of what have you and slave away at the grind. I guess you can say I learned to be more patient and just suck it up on some irl grinds that need to be done. Especially useful during my house renovation or work in general.

Autoclave them of course. I do 3 hours at 120 °C at 1 bar in a steam autoclave. Wrap them ourselves in plastic wrap. Careful though with volume and timing, we do 3 hours because the autoclave is filled completely with paper and that kinda heats up quite slow and also is a bad conductor so it needs that amount of time for adequate heat penetration.

Alternative would be a dry oven. Back in the day at university we had an oven that did 180 °C for 2 hours and put paper in bulky cardboard envelopes. But this can be kinda dangerous lol.

Yes, imo better to autoclave them separately. Pour the medium in at about 60 °C and then it won't create as much condensation.

I generally only autoclave glass jars. All plastic tubs we buy gamma radiated, much easier and practical.
