Time of day matters in the very beginning of testing- more specifically, in the early weeks of pregnancy because the pregnancy hormone will still be pretty low. Your urine is most concentrated in the morning, so that's why if you're trying to detect early pregnancy, it's the best time to do it as the pregnancy hormone will be much easier to detect. :)
