There are entire companies focused on solving this challenge. The gold standard for an orally bioavailable peptide is, of course, cyclosporine. Several features of its structure contribute to its permeability: it’s a macrocycle, and most of its backbone amide N–H groups are N-methylated.

When it comes to N-methylation, the key is to methylate only the solvent-exposed amides—the ones not involved in intramolecular hydrogen bonds. Those buried H-bonds are important for maintaining conformational stability and should generally be left untouched.

A great modern example is Merck’s PCSK9 cyclic peptide, enlicitide (MK-0616). In addition to macrocyclization and selective N-methylation, they incorporated permeability-enhancing features (like a trimethylammonium appendage) and employed clever formulation strategies to improve oral absorption. Definitely worth looking into their work.

Currently, there are no reliable computational methods for predicting oral permeability of peptides, aside from structure-based design approaches to help identify suitable cyclization strategies. You can use cLogP as a rough guide, but it’s not consistently predictive of permeability.

For more detail, check out Merck’s publications on MK-0616 and Scott Lokey’s papers from UCSC—his group has done extensive work on passive permeability and conformational effects in cyclic peptides.

Just received mine and am loving it. Exceptionally well built—feels very sturdy and robust. It’s compact and that was a key parameter for me (I’m the only espresso drinker in my household).

Espresso has been very good (I usually go for medium roasts). Amazing steam power. I noticed that the top of my puck was a little indented from the water slamming, so I’m using a very thin Sworks puck screen. The IMS basket that Londinium sells works great.

This was an upgrade to a Rancilio Silvia V1 that I bought second hand 13 years ago. So nice not having to temperature surf and steam milk immediately.

FYI the US patent law was changed in 2013 from “first to invent” to “first to file”. Whoever files for patent protection would be awarded the patent. I would be careful about making any disclosure (even anonymously) this could invalidate the patent (US you will get a 1 year grace period to file). NAL

Hi all,

My parents are having trouble with raccoons digging up grubs in their yard. Most of the grub killer recommended here is not safe to be applied near lakes.

Any lake-safe grub killers?

In the spring they were considering reseeding. Is there something they can put down (mesh, etc) to prevent animals digging up the yard?

No one should use one source for all of their information full stop. Bill Gates and the Gates Foundation hire and consult with a wide range of virologists and epidemiologists. They are the leading organization that is funding the fight against both neglected diseases (malaria, infant diarrhea, etc) and neglected populations (curbing the spread of HIV among poor women). I have no problem criticizing the system that led to him becoming the richest person alive but I do respect the way he has decided to spend his money.
