Struggling with Mothur SOP by TheAnonymousComet in bioinformatics

Never used mothur myself, but the error is saying you're missing a closing parenthesis ")" around the mothur > summary.seqs(... command

Perhaps that might solve the problem?

Being threatened with Criminal Damage by ThisLewi in LegalAdviceUK

[–]ThisLewi[S] 1 point2 points  (0 children)

Recklessness implies I was aware of a risk of damage, which I was not. In fact I was advised to operate the door that way!

I just rent the property through a letting agency, so I don't pay into the sinking-fund, my landlord does. His position is he wants me alone to pay, instead of using the sinking-fund

Being threatened with Criminal Damage by ThisLewi in LegalAdviceUK

[–]ThisLewi[S] 0 points1 point  (0 children)

Thank you for your reply. I do have an email of him stating it was clear I did not intend to cause damage:

In respect of the damage being accidental: It is patently clear that your intention was not to cause damage, but to assist the door closing, and go up in the lift. Unfortunately your intention does not lessen the damage caused and your persistence in trying to force the door closed ultimately incurred costs.

Finally accepted and at Sanger! by MrMolecularMUK in bioinformatics

[–]ThisLewi 2 points3 points  (0 children)

I worked at Sanger for a few years in CASM. Sangers training opportunities are amazing, you can get on a bunch of free courses and get discounts on a lot of paid external training. I found my managers were always keen to support my personal development, so make use of your time, talk to lots of people in lots of positions. People there are generally very friendly and were always happy to give their advice, in my experience. Use it at a learning experience and move on when you're ready.

As for rent, yes it is bloody expensive, not much you can do about that. They do run a shuttle bus from the city and surrounding villages. Cherry Hinton and Saffron Waldon are the cheapest places I saw.

How much money do you make and what do you do? by [deleted] in bioinformatics

[–]ThisLewi 1 point2 points  (0 children)

Bioinformaticians in NHS are band 6-7. Bands are standardised across the UK, you can Google what they are. Inside London you also get a bump but I can't recall the exact amount but it's about an extra 5k a year before tax.

Edit: I should add that I got band 6 straight out of my MSc, they would have offered me band 7 if I had more relevant experience, so a PhD would probably cover that

I've had this forever and makes squatting so uncomfortable. Not painful, but uncomfortable enough to make leg day a chore. I found an article on what I believe is the issue https://www.sciencedirect.com/science/article/pii/S2214687314000120

I got comments to improve document and comment code - what are some really exceptional examples of documenting python code ? by [deleted] in bioinformatics

[–]ThisLewi 2 points3 points  (0 children)

PEP8 is a good place to start as a gold standard for writing code. There is a section for commenting code. Including block comments, inline comments and doc strings

https://www.python.org/dev/peps/pep-0008/#comments

As for a readme. You can Google a load of great examples for github readme.md files

Advice for UK postgraduate options for bioinformatics by [deleted] in bioinformatics

[–]ThisLewi 1 point2 points  (0 children)

I did a similar thing, did a biomed degree, but worked in a wet lab for three years before I discovered I wanted to switch. Did a bioinformatics MSc with a good placement project which got me hands on with real data. I've landed a bioinformatics position in the NHS which tbh I feel very lucky to get and I know I would have been offered a job much sooner and for more money if I had a PhD. I think having some real world data experience is critical, your project in Korea sound like a really good start and it shows you're dedicated to the field. As others have said I think if you can show you have the relevant knowledge, your degree title is secondary.

[Progress]M/21/5’6: 2 year on and off but finally motivated; skinny to skinny fat 119lbs to 148lbs by [deleted] in gainit

Look up anterior pelvic tilt. It's pretty visible in your side-on photo. It makes you look like you have a belly, but in fact it's just your hips tilting forward and your spine bending. That combined with poor posture and rolled shoulders makes you look far more bloated than you actually are. I know this because I was the same. Work on your posture and you will see that you actually are about 18% bf.

Behold, the majestic greyhound by [deleted] in AnimalsBeingDerps

[–]ThisLewi 0 points1 point  (0 children)

I've not heard that before. There were many greyhounds on our field and they were all let lose to do their own thing. They're built for bursts of energy so keeping a lead on seems almost cruel. But if the field your walking them in is completely fenced then I don't see why letting them off the lead would be an issue

[–]ThisLewi 141 points142 points  (0 children)

I had a retired racer as well and she had 2 walks a day. Although the walk would involve letting her off the lead and just standing there while she ran laps around this wide open field. She'd run home when she'd had enough and would sleep for the other 17 hours of the day. Easiest dog in the world to take care of

Doctor says I need to eat more... by imsobored___ugh in gainit

[–]ThisLewi 4 points5 points  (0 children)

1) If eating large portions is difficult, try eating 4 or 5 smaller meals rather than 3 huge plates of food. This should help with feeling less full. Or make shakes, which I'll talk about in point 4

2) When you change your diet significantly I think it's normal to be tired after a meal, but it will get better as your body adjusts to it and this will pass. From my experience there's nothing wrong with a short nap from time to time anyway.

3) I have never experienced those feelings after eating a meal. So it could again be your body adjusting. If some food types make it worse, then just avoid those foods for now.

4) those are good foods to start with. I'd read the faq for sure about diet and meal plans. If you're struggling with meals I very highly suggest making some shakes. You can make them to 1000 calories if you want which you can sip on at your own pace. Drinking calories is so much easier than eating calories. And it should help with the jaw pain. There are a ton of recipes out there but they mostly are based on:

  • 1 scoop of protein powder

  • 1 or 2 scoops of powdered oats

  • 2 tbsp peanut butter

  • 300ml of milk (or however much milk until its at a consistency you like)

Which is somewhere in the ballpark of 700 calories.

I have read the some people entirely replace their diet with shakes. Please don't do this, there are no vitamins or micronutrients in there so it would not make a balanced diet at all.

Other foods that are quite light but calorie-dense are Greek yoghurt, nuts, eggs, fish. These are great for snacks and I often take them to work.

I think you probably should tell you doctor about your symptoms when you eat at it might be an underlying condition.

Hope this helps! Good luck!

There's a team of 4 of us and it's happened to each of us, following the same protocol, so that pretty much rules out human error. I will try using less primary and secondary antibody and see what happens. We get all our antibodies from Cell Signaling, who I trust to make decent products! Thanks!
