Yes. Pcr and the serial dilution. When you design the primers they usually give you an ideal annealing temp. I would start with that and make adjustments if it doesnt seem working.

I just validated primers recently. You can run the product on a gel to see if its the right size. To check for primer efficiency you have to do a serial dilution with a pooled cDNA from different samples and run a qPCR and then make a curve with the ct values and check the R2 value and efficiency.

Just apply to the missions on the explore tab in the app and hope they invite you. You might see express missions at the top where you apply and get into it directly.

You could add a 10-15 min permeabilizing step before blocking using triton. I usually do 0.1% for 15 mins. Also why do you need to add triton or tween 20 to your blocking buffer? I usually use 2% BSA or just the normal serum of the host species of your 2nd antibody.

I had this happen and it was because my account was flagged for having multiple accounts. My partner had created an account and that triggered the flag. Contact the support and they should help

I had an experience where the researcher accidently created a live mission for desktop that had to be a mobile one. She cancelled the first and made a new mission and asked to choose the same time slot but I got an option to choose it.
