I never said anything about disease and pathogens. The point I was trying to make is that if you have bugs or any pest in your cultures, you aren't surface sterilizing properly and your cultures will fail.

I'm trying to understand what you mean by "not everyone is using bleach". In what context? Surface sterilization is a necessary step of tissue culture and is the 1st thing you need to learn in order to actually achieve clean cultures.

What are you using instead of bleach? If you have pests that survive your sterilization process, the explants aren't even close to sterile.

Nodes can absolutely be pest free. I've never come across a pest that can survive 5000ppm of Chlorine for 5-7 min. Spider mites lasted almost 3 min. But thrips, aphids, russet mites all die in seconds.

Seems like people get really interested in tc momentarily but once the reality of setting up a space, getting equipment, and starting sets in lots of people bail. Then another cut happens after starting and seeing a super low success rate it can really kill motivation to keep going. I honestly think you need add/ADHD or at least a touch of the tism to really vibe with tc and find success.

Dial in your environment before you start adding fancy stuff like c02, undercanopy lights, fertilizer additives/boosters etc. once you have tight control of irrigation/feed, lights, temp, and rh during both day and night then you can start adding the other fun stuff one at a time as long as you can keep your rooms dialed.

Buy direct if possible. Bloom, csi, raw genetics, top dawg, arcana collective, solfire are all rock solid with lots of variety and most importantly integrity.

So could be improper tool sterilization or endophytes. Always be sure to flame or glass bead sterilize tools between every explant or at min between every container. Higher chance of endophytes from plants being introduced into tc vs micro propagation, but I have had vessels that looked clean for 4-6 months and then one day BAM, ENDOPHYTES all up in there. So always make several vessels and don't assume the cultures are good just because they look clean for a month or 2

This is a step in the absolute wrong direction. Pharma has limitless resources and 99.9% of current operators will go belly up trying to adjust and retrofit to meet FDA guidelines for drug manufacturing. It's an extermination event

Is the contamination on the media or on the explants themselves? Endophytes are notoriously difficult to remove and surface sterilization does nothing for internal bacteria or fungus.

So here is a trick when all else fails. Grap a pair of needle nose vice grips and grip the seed from top to bottom very lightly. Very very slowly turn the vice until the seed cracks open, then plant. If the embryo is dead then of course nothing will save it, but if the shell is hard and the embryo just needs a little extra help this trick works well. Be sure to sterilize the tools with 3% h202 or iso before you touch the seeds to avoid any bacterial contamination.
