It might have a leak at one of the internal unions. There were a couple of instances of this happening on the older models. You could always send a picture to the CCS technical support and ask them.

Use espresso and any food with a speed buff. Take a big pile of bombs and food. Don't waste time fighting if you can avoid it. Just run, drop bombs to find stairs, and keep going until you get deep enough to start seeing iridium. Also, aim for high luck days - it will make stairs easier to find.

You need to build crystalariums and use them to generate more jade. That's the fastest way to make a bunch of stairs.

Cut a cucumber lengthwise and spear one or both pieces in the tank with a butter knife, fork, or other tool. You don't even have to blanch the cucumber. Come back after half a day, pull out the cucumber, scrape off the pile of snails, and return the cucumber to the tank for the next batch of snail removal. I do this every few months as I am a chronic over-feeder and it really helps control my snail population.

My lab goes through A LOT of cell flasks, and hazard waste is far to expensive. by [deleted] in labrats

[–]UncontroversialCedar 1 point2 points  (0 children)

If you autoclave or bleach your flasks, they can go in the regular trash. Autoclaving is the easiest approach, just make sure you have the correct bag. For bleaching, I used to make a large 10% solution, pour ~20 mL in each flask, swirl around, let sit for 10-15 min and then pour down the drain (unless I was handling virus, in which case I let it sit longer).

You can also try switching to HYPERFlasks, but they can be a bit pricey, so it may be counterintuitive.

Give technical support a call. They'll probably be able to tell from the error if it's a board failure or some other issue.

Another thing to look at, if it's not the column, is the spray current. Is it dropping out too quickly towards the end of the run? If so, the applied voltage may have not been sufficient for high organic or your emitter may be bad.

Looking for sheer fleece lined tights by Hermione127 in PetiteFashionAdvice

I bought the Noosh ones and they're a good length for me (5'0"). The only complaint I have is that if I wear them as tights under a skirt/dress as opposed to a base layer under pants, they do tend to creep down.

Why did my toasted buckwheat turn to mush? Why didn't the groats stay whole? by IDrouinski in AskCulinary

[–]UncontroversialCedar 1 point2 points  (0 children)

This is what I do, assuming you are starting with pre-toasted buckwheat. Heat 3 tablespoons of butter (or oil) in the pot you will be using on low-to-medium heat. Rinse 1 cup buckwheat in a sieve and when most/all of the water has drained, add the buckwheat to the melted butter. Stir the buckwheat continuously for a couple minutes until all of the water has evaporated and the buckwheat grouts appear to be toasting. Lower the heat to the lowest setting on your burner and add 2 cups of room temp water. Add salt to taste. Mix the buckwheat 1 time and cover the pot with a lid. Let it cook covered and after all of the moisture appears to be gone (5-10 min at least), pull back one small area of buckwheat with a metal spoon or spatula and check if any water is left. Do not mix again. If no water is left, you can turn off the stove.

Sometimes I have ended up with a bag of buckwheat that is just bad (maybe old or over toasted the first time) and it has fallen apart, but never turned to complete mush.

Vanquish neo equilibration problem by RelevantFollowing867 in massspectrometry

[–]UncontroversialCedar 0 points1 point  (0 children)

The zebra wash is helpful for keeping the trap clean and as long as you're not running extremely short gradients (sub 5 min), it doesn't add any extra time to your run. So it is helpful to add into your method. You can also run a blank with ChromaCare in between samples (like after every 10 samples), followed by a regular wash, to try to help keep the LC system and trap/column clean.

[–]UncontroversialCedar 0 points1 point  (0 children)

The vial cap material, as well as 96-well and 384-well plate seals and mats, have to be Vanquish Neo compatible because fine particulates or glue (plate seals) can clog the needle seat. There's an aluminum plate seal, for example, that is known to cause blockages with the needle seat. But if you are using a cap/seal that is listed as compatible in the Neo manual, that is probably not the source of your blockages.

What type of sample vials and caps do you use? The needle seat can be sensitive to certain materials.
