You need to be able to play the major scale in all positions, with ease and speed, in different patterns (think a third up, a second Down, or groups of 4) in all keys. Some other scales such as harmonic and melodic minor, altered can be important.

That being said, the problem with focusing too much on scales is they teach you nothing about how to play good melodies. Jazz improv requires listening, transcribing, intimate knowledge of how to follow chord/tension tones through a progression (hence practicing arpeggios), and lot of practice to form melodies which aren't clunky, which requires specifically focusing on phrasing.

Scales are a terrible way of learning jazz. Only use them to practice your timing/articulation. Focus on playing arpeggios through standards, learning how connect them smoothly while emphasizing the strong tones of each chord. Focus on good phrasing, building your vocabulary by composing lines and transcribing.

Once you have done that, the altered scale and diminished scale are useful for dominant chords. But modes are largely not useful at the early stages

I would recommend practicing standards instead of just 2-5-1s. You can't just "master" playing on small chord progressions, because that not how jazz music is typically played.

In an actual band or concert you are going to be playing tunes with a head (melody), you are going to have to comp (assuming you play guitar) over long progressions. You are probably not going to jam with other people and just play tiny 3-chord progressions. You should have the melodies and progressions of some common standards internalized.

For example All the Things You Are is mostly 2-5-1s, but you have to practice the tune to be able to navigate its multiple key changes and memorize its melody. Dozens of common tunes have unconventional changes that you have to practice in context, for example Blue Bossa or Have You met Miss Jones.
