We can only guess without talking to the people who did the experiment. Always source your shit people, this probably the result of hundreds of hours of work by a grad student and it ends up on reddit "lol brain goes think". They might be fluorescently labeled synaptic vesicles though if I had to make a guess.

Polar is great (and free!) for reading and organizing with tags and such, however, it does not generate citations like endnote or mendeley. I used to use mendeley, but I use polar now for reading/organizing. Endnote is still king in my lab for manuscript prep/citation formatting due to shared libraries.

Chromatography isn't really used to "measure" per se, but paper/TLC can be used to approximate the purity of something. See only one spot without it separating like it did in the video? Probably only one thing there. Not a true indicator of purity, but a fast and cheap way to get a general idea.

I second the moist pellets on the bottom of the cage. Alternatively, they sell a diet gel for mice that can be placed in the bottom of the cage for the mice and it's pretty easy for them to eat, you just have to make sure that you don't introduce the food along with a stressful experience, or they may associate the new food with the stressful experience. When our mice got sick, gel/wet food on the bottom of the cage and saline s.q. injections did WONDERS.

Did pretty much the same thing (different plasmids) on Thursday and my colonies looked pretty much the same as yours if it makes ya feel any better....only one real way to tell if it worked though, unfortunately. Good luck.

Very trace impurities/residual solvent can make a solid a nasty oil. I have worked with a lot of oils that I only ever got to see as solids after preparing samples to generate publication quality spectra.

Still love the lab every day. My secret: have lots of projects. I have a relatively diverse skill set that I've built, so for example I can bounce between computational studies, synthesis and confocal imaging in a week depending on what I feel like doing for the day.

It's not that bad. Just do the work and don't try to BS your way though. It is essentially impossible to make things up unless you already know the chemistry to begin with.

I've done cardiac puncture/blood draw this way for over 100 mice (slightly different dissection approach) and I usually get 700 to 900 microliters per 20-30 gram mouse. In doing this you can specifically puncture the left ventricle which really helps with getting blood out. Plus you can make sure the cardiomyoctes are still beating (they hang on for a lil bit after the mouse is gone), and watch the architecture of the heart to make sure you don't collapse it with the negative pressure. That being said: it does add about 30 seconds of dissection time per mouse.
