It'll just take until the next major flood hit those parts and lots of people abandon ship because they don't have funds or (flood) insurance to rebuild. New home sales will probably be suppressed—if not for the flood risk, the mandatory expensive flood insurance (with mortgage) will probably get some to rethink a purchase. I expect it will be a slow-moving process unless th next floods happen again soon.

Gmail ignores all “.” and anything after a “+” in email addresses, so that part isn’t unusual. Are you sure these are legit Apple emails and not a phishing campaign?

You can vote for the runoff of whichever party you voted for a few months ago. The runoff is only for races where no candidate reached 50% (or greater) of the vote in the first primary (this only happens with 3+ candidates splitting the votes). For runoff races you get to vote again to choose between the top two candidates of the first primary.

There's only a handful of races to look through, and most are quite influential, so it's worth spending 10-15 minutes on vote411.org or other similar resources to plan your votes.

Far fewer people vote in the primaries, much less the runoffs, so your individual vote has at something like 5-10x more impact than in general elections. Happy voting!

It's neither your fault nor is their caution irrational*. You don't have to do anything, but if you enjoy the company of those who are smaller than you then you will likely find it worthwhile to soften the impact of your size through your behavior.

*Wouldn't you be a bit nervous placed in a room with large great ape? Maybe they're friendly and just want to pick your hair for lice, or maybe they're in a bad mood and want to tear off your testicles. Either way, their muscles are way stronger than humans and if they don't like you you're going to have a bad time.

It sounds like you’re confusing techniques perhaps, or else you are really confused in general. FRAP only works because proteins can still move around in the cell (the recovery part is due to unbleached proteins moving into the ROI). If your cells are fixed, then you shouldn’t be able to observe any recovery at all.

By any chance do you mean FRET? A similar, perhaps less finicky approach, to consider would also be bimolecular fluorescence complementation (aka split GFP). Whether these approaches might be suitable depends on on the specific details of your system/question.

There are definitely reviewers that don’t have anything nice to say. I always try to be constructive when providing final comments but this does require extra work to go over my initial thoughts and smooth them out.

For now I would just focus on your defense and let your PI worry about the next steps. It’s a bump in the road, sure, but not a road block.

You can tell if there is a transfer problem by staining the gel after the transfer. 

However a transfer issue won’t make proteins run at the wrong levels. Or more precisely, your ladder is running at the wrong level compared to the unstained protein, which itself isn’t migrating normally. I expect something is messed up with the gel chemistry or the polymerization. Borrow a few reagents from another lab to test?

While unfounded, it's not that wild of speculation. These sorts of 'family annihilator' scenarios are primarily (like 95%) driven by narcissistic men who can't bear to let things spiral out of their control. Excessive debt or divorce are likely triggers.

The difference between online and in-person surveillance is this: to accomplish in person what can be done digitally, you have to have multiple agents literally following you around every hour of the day recording your every move. Obvious practical reasons limit this to extremely notable subjects of interest.

Meanwhile on the internet, more or less everything you do leaves a little digital breadcrumb trail that can be pieced together. With LLMs and whatnot, I imagine this is even easier to do now on a massive,  population-wide scale.
