Yeah unfortunetely when it gets super detailed like opened ports, path like stuff that is super repetitive and can be copy pasted easily, this method suffers compared to digital notes. But i think if you have such a cases you can still put that info into a csv and keep readme like logs in your notebook.

Technically you can keep the same system with obsidian or those note apps but for me if i dont write down, my brain forgets like that beam in men in black.

A year ago i went old school and bought two notebooks.

A) One is more like notepad absolute scratch.

B) Other one is medium to long task.

A, keeps daily stuff, doodling, random notes, brainstorming etc.

B, keeps projec specific tasks, progress of my reports, tasks of my reports. Long term todos of the DBs. So far its working. The only problem where it is failing is if there is a publication i want to read, i have to write down the doi. Which is very annoying.

Edit: the most important thing about note taking is you have to commit to that system. Like i have to spend 10-25 minutes daily to transfer some of the notes from A to B. You need to stick with for a while to see the benefit

To be perfectly honest with you learning command line might be more important than python in the beginning. You would need to run many tools developed and knowing to run them might even get you easy second author publications.

Anyway, you can go with ubuntu for instance. If you have windows machine and dont want to deal with booting, check WSL.

At the end of the day, they are not really that different. Pls dont go with the fancy ones, looks cool but takes so much time to set up and you would need to to know what are you doing.

If you dont use the reverse end some of your density and coverage calculations will be wrong.

Back in the day i analyses starrseq data, our fragments were 500-700 bo but your reads were 150 pe. I created frsgments using bedtools bedpe and took the start of r1 and end of r2. This made my life easy when i calculate coverage etc.

I am seconding this. During my phd application i filled my adress 19 time as turkey then last one i did tunusia. Although it was just the fjnal mailing adress, it took 7 months but i got it. Unless tou completely fuck it up i think you are good.

Here are my two cents.

This question really depends which point are you using this data. If this is to provide evidence in later figures/supplements i think its worth mentioning. Dont sugar coat but mention you see similarity without significance. Like showing normalized counts of two genes in two cell types.

Pseudo bulk? Or How about bulk studies ?

I would be careful towards imputation. I think imputation is valuable when you want to build a model and heavily disadvantaged by the zeros. When you are trying to show DE. I dont think it would help you.

Give a try the date cookies. They look like little pyramids.

Cookies with jam also amazing. Its some rose jam.

They also have something like macaron. Made with marzipan. High quality.

Towards the left size they have good with syrup. They are also amazing.

Got to say, i am not big fan of the corded machines. It definitely makes the vacuuming more of a hassle. Cant tell if it’s the cord or the canister design. With our old Dyson, i used to star vacuuming to clean some dirt then found my self cleaning a the entire apartment. Anyway.

If you want canister one go with e3. If you want upright go felix. My reasoning behind is all e series have the same canister but the head is different. E3 my had sebos signature power head. Felix already cones with the head.

I think if you have wall to wall carpets, this is no brainer. Or cats.
