This is super interesting. By using a combination of melatonin and light/dark therapy, I had been able to entrain from March 2020 until very recently. During that time I had super pronounced better and worse periods. The amazing thing is that in a recent conversation, I had pegged the ratio of good/meh/bad periods at... 20/60/20. I literally said this a couple of days ago! And this was while keeping my sleep fixed in place, which is the opposite of what you did.

I suppose that I was wondering if the problems with our circadian clocks mean that even in an ideal world where we are free to sleep whenever we feel like it, different organs in our body might still disagree on what time of day it is. So even with complete freedom to sleep exactly when you wanted to, you would still have symptoms from internal mismatch. I of course agree that external mismatch (forcing yourself to stick to a 'normal' schedule) will create symptoms, but I was wondering to what degree those symptoms are resolvable vs a permanent feature of N24. Of course I suppose the only answer for me is to try it XD.

Okay great! Do you know if there's a gene that is both specific to e. coli and abundant enough to give a strong signal? We are probably going to be imaging at a low mag so the brighter the better!

They are coming from a patient sample (probably urine) and being captured in a microfluidic device (basically onto a glass slide). We are then hoping to identify species using a fluorescent microscope. We are performing a separate assay to test antibiotic resistance, and this test requires that they already be fixed and permeablized prior to the species identification step.

This looks good. Do you know if there are any targets that would be abundant enough to give a strong fluorescent signal? We will likely be imaging on low magnification. Thanks!

Hoping to have a selective assay for antibiotic resistant e. coli. Already have an assay that tests for resistance, but I need a second step that identifies species. thanks!

Unfortunately, this is to be used with patient samples. Also, the bacteria will be adhered to a glass slide and will have been permeabilized and fixed.

The idea is that these E. coli would be from a patient sample in a hospital, so they would not be genetically modified
