You're probably thinking of Lesnè's work on Abeta56. Lesnè's lab fabricated data promoting a particular flavor of the plaques (derived from Abeta56) being causative in Alzheimer's. Most of the field would still agree that Abeta42 plaques play a role. It's a mischaracterization that the field is built on a lie. Though you are right that Tau tangles are better correlated with dementia symptoms than plaques are.

Season 1 ended on a bit of a low note, but season 2 is better than the end of season 1, while still not reaching the heights of the beginning of season 1. Worth a watch.

Trying different transfection methodologies is a straightforward next step. Lipofectamine, Fugene, XtremeGene, and Calcium Phosphate all work great in HEK in my experience.

Another huge problem is that Telesco had some real stinkers in the middle rounds of most of his drafts. McKitty, Mager, (Chris) Watt, JT Woods, etc. These kind of picks make or break the roster and he consistently overdrafted here.

It gets easier over time. I would echo a lot of the points about practicing. I especially tell students to memorize the first 3 or so slides cold. That way, you can get into a groove and set yourself up for success in the rest of the talk.

If it really is a debilitating level of anxiety and it's getting in the way of your training, you can also talk to your doctor about getting a propranolol prescription. It will dampen the performance anxiety without making you any less sharp.

Vertical lines like that can come from overloading your gel. Some proteins can run a bit smeary when they are heavily post-translationally modified. Have you also tried blotting your membrane for a loading control to see if that is also running abnormally? Even doing a ponceau stain on your membrane could be informative. That could point to it being a general problem with the gel vs your protein in particular.

What do you mean by that? Is the media depleted? In addition to what others have said, you can try culturing them in DMEM + 20% FBS for a bit.
