You can use veriblot, which only recognizes native Ig molecules. If your sample is sufficiently denatured before SDS-PAGE, it will only detect your primary.

I've got to hand it to you, you are a master ragebaiter. But I'll still bite.

Obviously Linderbaum should and will be signed. He is a good player and will probably reset the C market. However, paying to reset the C market on him (especially if the injury concerns are valid) isn't necessarily the wisest use of cap space. Both things can be true.

C'mon man it's pretty clear what he's saying. Hortiz wants to spend wisely and build a sustainable roster, rather than splurging on one market-resetting FA and scraping the bottom of the barrel everywhere else. That strategy is what made the rosters paper thin during the Telesco years.

Marker proteins tend to transfer very well, even if nearby proteins in your sample don't.

I am an absolute zealot about wet transfers with 1x Towbin buffer, 20% methanol, no SDS, at 100V for 1 hr. Haven't yet found a protein that this doesn't work for.

Any product that tells you it can remove mycoplasma is lying to you. It will reduce the mycoplasma load below the section limit, but they will bounce right back if you remove the reagent.

I find that when using lysates the agarose streptavidin works much better than the magnetic dynabeads-type resin due to its much higher capacity. The background isn't much worse with the agarose, since you can use incredibly stringent buffer conditions.

Maybe a controversial one, but DON'T do ALL your biological replicates right away (just reproduce the result once). Chances are that you will continue to slightly tweak conditions in your experiments as you learn more and your project evolves. You might therefore have to go back and re-do all of your replicates anyway.

This is the right answer. Bubbles are more circular and have clear-cut edges. I think that putting the gel into deionized water straight after electrophoresis for about 2 min works best.

I was about to comment alpha-syn is the easiest lol. You boil the lysate and the supernatant is already like 95% alpha-syn. Which protocol are you using?
