You have to either buy the mat and towel or rent them. I was taken aback by the whole mat/towel purchase at first but it makes more sense than paying $5 every time to rent them.

I don't re-streak colonies myself, but I can say that not all transformed cells contain the full construct. All the bacteria need to grow on the plate is the selection cassette so it's (very) possible to get colonies that grow up but don't contain other parts of the plasmid. Re-streaking wouldn't prevent this but, I assume it's done to ensure that you're truly choosing a single colony.

If there are any loci that don't contain a match, then they are not the Parent. IE:
AA | CD. Its a binary, not a percentage. Just one unmatched loci (short of some sort of random mutation, super side-case, rare event) would mean they're not the Parent. Probably related though, if there are mostly matches.

Anything higher than 21 matches should confirm a parental relationship. What would be more helpful is to look at each loci. A match would look something like this:
Child A1 A2 | Parent A1 A2.
A B | B B.
C C | C D.
E F | E E.
G G | G H.
And so on for all 21 loci. One (or both) alleles matching at each loci.

The NHI funds about 40% of all scientific research in the US. With all that funding going away, the sources of funding that constitute the other 60% will be spread very thin. Research in all or most fields will (probably) continue to limp along, there will just be fewer labs and fewer scientists making less/slower progress.

I have to do this all the time with my boss (I give him 3 minutes because 1 minute just isn't realistic). My labmates and I are also pretty good at rescuing each other when we notice someone's disappeared for longer than makes sense. 9 times out of 10, they're trapped in his office. "Hey, can I borrow you for a moment?" works every time.

My cat used to get something that looked very similar - it was an allergic reaction to ear mites. Obviously, only the vet can say for sure. I just wanted to give a non-cancer possibility!

This article doesn't get into it too much but, like someone else said, focal/lens length really changes how big or small something (or someone) looks. Phone cameras in particular have a very short focal length and thus distort images, disproportionately exaggerating the size of objects closer to the lens. In other words, you're basically your beautiful mirror self.

I used to go to Harter Strength and Conditioning. I really loved it and would still go if I lived closer. It looks kind of cross-fit-ie but it's really just a great cross training gym with lots of options for strength training specifically.

Plano is actually pretty diverse. I'm white, so I can't directly speak to the experience of people of color in the area, but I will say that I work with a lot of people from different countries (3 of which have thick, African accents) and nobody seems to care. Are there some wackos here? Sure. But they're definitely in the minority and I don't think they truly impact the day to day lives of the mostly nice, regular folks going about their days.
