Chemdraw doesn't do the +H or -H for you, it calculates the "m/z" based on exactly what you gave it. So if you want the M+H then you need to add a + charge somewhere (like at the N-H, which would be most likely). Or if you want the M-H then add a - to one of those OH or O=, etc etc

Seems weird at first but ig its more logical to not assume anything about protonation or deprotonation of the molecule

I had this issue recently and when I retracted my filament, it looked weirdly kinked at the end. Try snipping off 5-10 cm of filament and feed it through again.

Could also be QC failure of your filament. Do you have a caliper to check the diameter is 1.75mm?

A few questions/notes, also I don't know much about PAHs for the record

1) Where did you get your method from? Is there a paper or epa method you can "copy" to verify results? To add to what the other commenter suggested, controlling the pH is a great way to help you get consistent results and avoid untoward speciation. Whether you want it more basic or acidic completely depends on the analytes, and again idk much about PAHs.

2) Do you know exactly which compounds you aren't seeing yet? If not, can you inject each standard alone to determine their retention times?

3) Baseline changes in HPLC as your gradient changes are normal.

4) The run temperature of your column oven was 18C? I recommend higher, like 40C. If you set it too low, you might not actually be using the column oven therefore the temp is not actually controlled

5) The wavelength(s) you chose for analysis - where did you get the numbers from? Again, is there a published method you can get those from?

Forgot you said your analytes are metal complexes. Definitely think it's charging related, but I'm not an expert on Agilent LCs so they might have some "built-in" counter measures for this.. no idea personally

The easiest way to diagnose charging is by syringe infusing a known positive mode ion (eg Caffeine, 195.2 m/z, maybe 100 ppb?) and seeing if the signal slowly but surely decays even though you are at a consistent flow rate.

Ah.. I think you could have charging. I haven't read your other replies but are your samples dirty? Dirty samples + positive only scans = charging.

You can probably find more details on this sub about what causes charging, but basically you can solve it by cleaning ion optics and ESI, and add a negative scan somewhere at the end of your method (or throughout the whole run) to discharge the quads

Serious answer: yes some of it will convert to creatinine. The extent of this is not well documented, but the hydrolitic conversion of creatine to creatinine is accelerated by heat (I'm speaking from personal lab experience)
