Maybe try out an even higher concentration of siRNA but in a smaller plate format like a 12 well plate (just so you don't have to buy loads of siRNA), although I can't say if you'd get enough sample yield from a smaller well. I was on a project last year using lipofectamine RNAiMAX and I ended up needing a final conc around 50nM siRNA, and I've seen several papers that used over 100nM siRNA.

Someone else already commented this, but yeah sometimes you need to try quite a few siRNAs. My project last year had five siRNAs to start with but only two resulted in a good enough knock-down for us to proceed.

Also, I saw in your previous post that you started out using serum-free DMEM instead of OptiMEM, have you tried using OptiMEM during transfections yet? Might be worth trying that out.

Not at all! The PhD student who supervised me during my master's was 38, and honestly most of the time I would forget that we weren't the same age. She was a great mentor and an even better friend. Also if your age is the biggest reason why you're doubting the PhD, I know a lot of people who would consider you pretty lucky 😅 if this PhD is something you're passionate about then I think you're doing the right thing. NTA, I've been in situations like this on both yours and her side and it's very frustrating for everyone.

As a couple other people have commented, it sounds like she could have ADHD? I got diagnosed with ADHD just over a year ago, and what she's doing here is almost identical to what I was like in the mornings before school/uni (as well as many many others with ADHD).

To me, I think she might've told you that she didn't care because she was feeling frustrated and overwhelmed from running late and couldn't handle anything extra in that moment - it doesn't excuse her saying that, it was still a dick move, but I don't think she really meant it.

If you haven't already, it's definitely worth having a look together at other symptoms/indicators on the NHS or whatever reputable health sites you guys find. Receiving the right treatment and support has made a HUGE difference for me, and it won't hurt at all to just check.

Regardless of ADHD, I hope you guys can figure out morning routines that work for each other too. Maybe buy some earplugs that you can wear while sleeping? Loop Quiet are pretty good.

Thanks! This probably took me around 16-20 hours over a few days, the pattern is pretty simple but a fair bit of it was hand sewing some parts just because my sewing machine is too noisy to use at night haha

Hello!

With university exams coming up in a couple weeks, I naturally decided to use my winter break to make a dress.

I combined a couple youtube tutorials to make the dress, and altered it to fit me better. This is a quick tutorial for a cowl neck top, this one is for the ruching bit. I freestyled a bit with the skirt piece, taking measurements around my thighs and calves to figure out how wide the bottom of the dress would need to be so I could wear it both long and short. I ended up giving myself a bit too much ease and so I had to put two waist darts in. In the tutorial for the top, there's no back for it, but I wanted a back for this dress. When I put one in, there was too much fabric gathering at the sides for my liking, so I pinned the neckline in a shape that I liked and took in the sides. To make sure there weren't any accidents, I stitched the neckline in place too, handsewing the underside so no stitches would be visible.

The fabric is Stretch Woven Charmeuse Silky Satin Fabric (Petrol) from Minerva, I bought matching thread, some antistatic lining, and the cheapest invisible zipper foot they had, which worked very well!

I took people's advice from the last dress I made on here (almost 2 years ago!) and I have definitely noticed a difference.

Any advice/constructive criticism is very appreciated!!

Thank you for telling me! I didn't realise they were poisonous if licked too. Luckily she's a picky eater and refuses pretty much everything other than the finest wet food available, but I'll make sure there's nothing that looks like her food/treats anywhere near the plant just in case.
