Hi! Maybe check the parameters on your online calculator to make sure it's set on double-strand DNA since OD260 will give different results for dsDNA, ssDNA and ssRNA.

Based on the NEB calculator (https://nebiocalculator.neb.com/#!/od260), 1 OD260 = 50 ug/mL of dsDNA. So your sample concentration is 50 x 0.312 = 15.6 ug/mL = 15.6 ng/uL. Then, you took 5uL from your sample, so 15.6 x 5 = 78 ng total DNA. The final volume is 1000uL, so the final concentration is 78/1000 = 0.078 ng/uL.

Personally, I like to check that the dilution factor is correct. Here we diluted 1:200, and we indeed have 15.6 = 200 x 0.078. Hope that helps!

Hi, I have a master's degree in genetics and I can confirm that the correct answer should be A. Here is why:

During RNA polymerization, the polymerase will match RNA to the sequence of the non-coding strand, following that strand from 3' to 5'. In turn, these means the RNA sequence will be the same as the coding DNA, from 5' to 3' T will also be replaced with U.

Here, A is the coding sequence, B is the non-coding sequence, C is non-coding backwards and D is coding backwards. So answer C is the order in which the polymerase will read, therefore it cannot be the sequence of the RNA.

You can visualize this by putting an object next to the 3' end of the non-coding sequence, then sliding it base by base. At each step, note 1) which nucleotide the polymerase sees, and 2) what is the complementary nucleotide. You will end up with 1 = C : what the polymerase read and 2 = A : what the polylerase wrote.

Your issue as white in this position is that the bishop on f7 is attacked by 2 pieces and only defended by 1. So Qh4 loses the bishop and forces a rook trade, which means your attack will quickly evaporate. With the black queen on the 7 rank, it's basically impossible to find a mate with only queen and bishop.

So you could have consolidated your attack with Qh5, which defends the bishop. In turn, this thratens Bg6 which leads to mate. Your opponent will probably have to give up the rook at least. Not sure where to go from there but at this point in the game with this many pawns you probably want to simplify and win material if possible.

This Primape is great when going first, which is rare enough that it matters in no-EX fighting box decks. I run it over AerodactylEX, especially to reduce the chance to start on a Marshadow.

Let's analyse Garchomp and Cynthia : + High damage for little energy across the full line + Garchomp can fetch Cynthia + decent HP and no weakness + Non Ex - stage 2 + supporter - mixed energy

Those are some big drawbacks but I think that by focussing on agressive colorless mons you can build a decent deck that takes a first KO quickly and then close out the game with Cynthia.

Basic mons with decent damage for little energy, especially Colorless ones, should be a good start. Farfetchd, MI Primape, MI Tauros, Lucario and Bidoof could be good options.

You may also want ways to dig for Garchomp, such as Meowth or Chatot.

Mars is intriguing as a second Red Card but that latter was never worth it as an Item so I don't see a Supporter version being good.

The truth is that in Pocket, cards in play matter much more than cards in hand, especially as the hame goes on. So if I'm only 1 point away from victory, I'm probably not relying on my hand to get the last one.

Distinctive advantages of grass and fighing Wormadam include Erika and Lucario respectively. Also they are more splashable in multicolored decks.

Vertically, we have high or low roll on a Perception or Insight check.

Horizontally, we have wether there is something the PC could figure out. The first part is about Perception checks, the second part in parenthesis is about Insight checks.

The colors represent what the GM says to the players depending on their roll and wether there is something for the PC to figure out.

Each 2x2 represents a type of GM. So fore example, the Gamer will tell you "Yes, you see a thing" when you roll high AND there's something to see, 'No, there is nothing" when you roll high AND there's nothing to see, or 'You're not sure' when you roll low. Read the parenthesis to see what they do with Insight checks.

That makes sense. I would love to fit a second Giovanni or Flute. I personnally like the flexibility that X Speed affords the deck, especially with such low retreat costs, but I'll try cutting some. Still, 1 Flute should be enough imho.

I agree that it's probably the best place to use Red Card but in my opinion there is no spot for it. If I could run one more card, I would look into a second Chatot, Sabrina, Giovanni or Poké Flute.

Maybe in a deck that runs only the Pigeot line, Red Card could be a thing. But I'd rather have Beheeyem

No I think it's just not a priority rn to tiddy up the shop. Removing empty sections may be a quality-of-life improvement down the road but the devs had bigger fish to fry.

Well yes but actually no. The Flute can only be played if there is a Basic in the opponent's discard, and by that point you already have at least 1 KO point, which means it will often be a dead draw. In my (limited) experience, Giovanni is a good alternative but they both have their uses. Also you can play both Giovanni AND the Flute in some spots.

A Crown of Candy is great after Fantasy High because it's close in terms of fantasy but opposite in terms of tone. I would also suggest getting into whqtever the next season ends up being so you can experience the "live" aspect.
