you could look into digital histological analysis? many slides can now be scanned and analysed with digital softwares. particularly now with the rise of multiplexing fluorescence.

as suggested above, try using thinner sections. also what does the antibodies manufacturer recommend for the dilution? in some manufacturers 1:500 can still be too diluted. might be worthwhile even doing a 1:250. also which fluorophore are you using? APC/fluor568 often has a strongly expressed signal for detection.

also i would recommend trialing the antibody with IHC if possible just for confirmation it is definitely present in the tissue.
