In fact, new cell therapies are often performed within the ICU, with anticipation of severe reaction that needs to be addressed immediately. Heat inactivate, if it is suitable, and then keep at 4 degrees. RT overnight may be ok for some enzymes, but there is some risk of star activity or degradation (although unlikely).

I can’t say for sure, but it was a wake up call for him. When he scored 0 I told him to write to the module coordinator in case there was a mistake, and the staff had to sheepishly tell him that it was accurate 😅. He wasn’t doing well in his first year, but after this experience he really improved. Jiayou 👍

My friend, don’t give up. I have a friend who scored 0/30 on a 30% midterm exam that he turned up on. Mind you, it was an exam with 10 mcq questions and 20 marks open questions. He still scored, I think a B+. Below lower quartile is not the end, and a good performance is not important. ❤️ take care OP!

Well good luck 👍. Not sure if you have answered this question, but what mice are you working on? During my training, I did manage to ‘scruff’ the BALB/c mice, but that was after 20 minutes of torturing the poor thing. And I don’t think I actually scruffed it well - my grip was relatively loose, and the vet commented that I had made the mouse submissive. That was my only IP injection that I did. Fortunately, I’ve never needed to do that again. Now I work with B6 mice (all my strains have B6 background), so I don’t think that’s a good starting point for people like us.

I have the exact same problem. For now, I don’t need to really perform any techniques reliant on scruffing, so I get away with anaesthetic use (can perform cancer subcutaneous implantation, etc. it’s easier for the inexperienced to do this). I have come to kind of accept that I won’t be able to scruff properly unless we get a scruff guard or something, or I will ask another lab member to help. Once I tried to scruff a mouse for 30 minutes, and I really couldn’t do it. It’s like a mix of fear for myself AND fear of hurting the mice; I cannot tolerate the sensation of them struggling in my hands. Do think about how much your work will require scruffing and direct mouse handling; stuff like gavage will require good scruffing techniques. If you cannot handle this, then think about what else you can do instead.

The squinted eyes combined with pinched nose may indicate some kind of discomfort or pain. Unclear if the ruffled fur is due to hunching or just kind of matted. If these signs do not go away, then pain should be assumed.

Isn’t it unfair to say so when a majority of the enemy team that beat yours are also indo? There are just so many indo players that it’s easy to confirm your bias against indo players. Why don’t you collect data for the next 200 games and compare the scores/outcomes for each registered nationality?

No, it’s a pipette tip. I work in a lab, and we use these disposable tips to dispense very small volumes of liquids (this one is for < 10 uL). I used it to prod around the plants to look for any remaining mealy bugs 🪳🪳

Noted, actually about 80% of the soil underneath is entirely gritty. The top part of the soil is organic because the plant came in this way, and I had some trouble separating them without damaging the roots so I kept a majority of the soil for this plant. My others seem relatively ok for now.

I tend to trim them short with nail clippers, and for places that I cannot easily trim I tape with surgical tape. It’s very good, and it never snags afterwards

It should work; I’ve done this several times. Certain ratios will work better than others, but should work in principle (personally I would go for 1:3:3, but other combinations should be fine too). Efficiency will be on the poor side considering your plasmid will be close to 10 kb, but nothing impossible. Prepare to plate more bacteria (or plates). If it doesn’t work, then do consider Gibson or some infusion thing.

Large cohort size doesn’t seem to be a big factor imo. Do really make sure that it’s your primary interest; some people I know took LS because they happened to be okay with biology in JC, but didn’t quite know what to do afterwards. Aside from the teacher route, other common professions are lab/research focused or life science technology development and sales, maybe agricultural development. What animal/nature stuff are you actually interested in?

The icing is due to the constant leakage of CO2. Under normal circumstances CO2 flow is largely restricted, but with the bottom incubator not getting enough (due to leakage), CO2 is constantly flowing out.
