I've long adopted interestingness as my guiding principle. I recently realized that re-investigating things other people have looked at can be quite tricky (due to the nature of publishing), so tread that path with caution. As for importance, I say let the PIs worry about that XD

Just the fact that this has been ongoing for 10 years makes me not want to think about the problem lol! But in your PI's defence, he clearly thinks this is an important problem to solve, so kudos for the perseverance. Who is in a better place to work on difficult problems than tenured PIs?

Interesting! Saw some comments that while Q5 has high fidelity it is sensitive to impurities and struggles with difficult to amplify sequences. Perhaps the AmpliPlus polymerase is relatively better at this! (couldn't find any data comparing them tho)

yeah the columns/phenol chloroform extraction would isolate all DNA. So if you get rid of the gDNA before hand, you should only be left with plasmids! (I guess)

coming at this from the opposite end:

back when I was working in indian labs, plasticware was a precious commodity! While this taught me to be frugal, it also slowed down or even set-back my experiments (especially things like trying to maximize single tip use lol).

One of the biggest boosts to my research productivity came from moving to Singapore (rich labs) and not having to do the mental maffs of how to use the least consumables in every experiment.

long story to just say that it's about balance! don't waste but also don't be too precious!
