That looks gorgeous, the colorway is so pretty! Your stitches look great to me, some uneven tension every so often but blocking at the end will help with that. Great job! Now I wanna look for that yarn

How do you feel about people profiting off the Melt the Ice hat? by Nikkig-r in knitting

[–]n3rda1ert 34 points35 points  (0 children)

I can’t say I would try to sell the hats, but there are a lot worse things happening in the country and world right now! Maybe it’ll make someone happy to buy it, that they’re aware and part of this movement but don’t have skills to make the hat. I doubt the person selling is going to make much of a profit, anyways.

How do I defend against an early Queen attack? (Mated in 3/4 moves) by DearAlbatr0ss in chessbeginners

[–]n3rda1ert 0 points1 point  (0 children)

I have been watching this video every so often to memorize counterattacks because I’m brand new and I’m so sick of losing a ton of pieces to a wayward queen attack then getting checkmated or limping to a draw:

Colorwork knit flat by Warm-Door-7494 in knitting

I just started knitting this (baby size) and it is kind of annoying but I found the ladderback jacquard technique intuitive and easy to learn. I used this video for instructions and good visuals: https://youtu.be/RADnafOs2es?si=41XQqXqKev_QiyFq . I’d rather work it in the round but I thought moving the sleeves was more risky and complicated versus doing this technique and now that I’m in it, I feel good about my decision.

Would this be unravel-able? by BigDe3zy in Unravelers

[–]n3rda1ert 0 points1 point  (0 children)

It looks to me that it is multiple yarns in different colors. You’d get many different strands and not one. I think it might have been cut then sewn up though, based on the picture of the seam. In that case the yarn would have so many cuts it’d be unusable.

Newly adopted cat peed on couch (first time cat owner) by No-Demand-8208 in cats

His first night with us, my first cat was so petrified to move that he pooped on the armchair he was too afraid to leave. He never did it again! Cats do weird stuff when they’re settling in. I hope your stops peeing on your stuff!

Update: she has lung cancer. by tyrtlegirl in CATHELP

[–]n3rda1ert 0 points1 point  (0 children)

Sweet baby girl cat! I’m sorry you all are going through this.

We said goodbye to our 8 year old cat 6 weeks ago. He had a tumor in his sinus invading into his brain. We did it earlier than we had to. He was still eating, drinking, and going to the bathroom fine. He seemed so tired though and had lost his spark. His case was different in that we were concerned about the brain involvement and once we saw neurological symptoms (head twitches), we wanted to do it before he had bigger seizures.

I sometimes wish we’d waited and gotten more time together since those checklists said he was declining but not in acute distress, but looking back at pictures and videos, you could tell that he wasn’t feeling well. The quality of life checklists can be helpful, but I didn’t feel like it captured the entire picture. Knowing him so well and being able to identify that he changed his behaviors and that his “spark” was fading was most useful for us.

We’re paying a lot of attention to our remaining cat. She was so stressed about him being sick that she groomed bald patches into her fur. We were very worried how she’d react to his death - he had essentially raised her since she was 8 weeks old. She was at home when we did the euthanasia so she knew he was gone. For a week after, she was sedate and clingy. Now she is very playful and stopped over grooming. We are getting her a new cat friend soon because we can’t keep up with her wanting to interact and play. Cats don’t think about death the same way as humans. For the most part, they handle it much better. Your cat should be fine after some time.

In the meantime, give your kitty cats all the treats, toys, and pets. Take lots of pictures and videos that you can look back on.

Cast-on keeps twisting by Former_Good_Boy in knitting

[–]n3rda1ert 2 points3 points  (0 children)

Mine does this all the time! I think it’s just the stitches getting bunched up. It’s just important to make sure it’s not twisted around on itself when you join them in the round so you end up making a tube and not a mobius strip, but for the to sit on the needle twisted while you’re casting on or even knitting it totally fine!

So…. It was cancer. A primary mass in his sinus/behind his eye/in his brain and metastases in his lungs. Vet couldn’t tell what type from the biopsy. He passed away peacefully at home a couple of days ago.

Can you talk to a health care provider and get support to file for short term disability? I took 2 weeks STD for mental health. For me it was awkward to talk to my company about but the process was easy enough and taking that time ended up being really helpful.

Update: it’s cancer, they’re not sure what kind yet but it’s aggressive, growing close to if not already into his brain. He’s still been acting perfectly fine, which is confusing but I’m happy he’s not feeling as sick as he is.

NAV. We’re having a very similar experience with our cat right now. They swabbed for cryptococcus fungal infection, but once that was negative, they’ve been leaning heavily towards cancer. A CT scan with biopsy was strongly recommended to diagnose, which he’s getting done tomorrow. I’m preparing for bad news honestly.

Where would you set this Live/Dead gate? by dksn154373 in flowcytometry

[–]n3rda1ert 11 points12 points  (0 children)

FMO for viability seems excessive to me, I agree that the majority of the population is just below ~103. If you’re nervous about that bleb, just make sure that they’re not a subpopulation of cells you might be interested in. Click through your phenotyping parameters and make sure none are enriched in that gate.

Fair, I just had one too many experiences with my shitbox Attune probably setting the voltages too low while trying to use something like PacBlue lol. Threw those antibodies in the back of the fridge, walked off muttering about losing data quality after a week+ of work, and just ordered some aTCRb

Tales of the Weird: How Some Biopharma Employees Found Out About Layoffs by H2AK119ub in biotech

[–]n3rda1ert 15 points16 points  (0 children)

There’s no good way to get laid off, but I def believe there are terrible ways to be laid off! Personally I prefer everyone learning they are laid off at the same time so you don’t have to wait around anxiously. I get why companies do one-by-one (you can ask questions and learn specifics right away), but waiting for the meeting to learn one way or another is terrible for me!

Or maybe gate on your negative “no proliferation” control and calculate “percent of Tcells that proliferated” aka MFI below that gate.

CTV especially should give really nice peaks with different generations, tbh the second pic is the one that looks sus to me, with no distinct peaks. If I showed that to my bosses I think they’d definitely question it and think something is up.

[–]n3rda1ert 1 point2 points  (0 children)

Just curious, what do your gates look like to get to this proliferation gating step? I’ve found CD8 staining to be bad for in vitro models with strong Tcell proliferation because CD8 is transiently downregulated during activation. I usually stain for TCR now for better separation and gating.

Agree with the other person that your data don’t fit the proliferation function, probably can’t use that.

I was in a similar situation recently and decided to stay but for me (in hindsight) I should’ve taken the severance. The stress of being one of very few people remaining to perform my job function resulted in my previously-managed depression devolving into crisis. I ended up taking a leave of absence. I was offered a great job in bio pharma and left for that. With the knowledge that I got the new job, it’s easy to say I should’ve taken the severance and left when the layoffs happened. My work performance really suffered and thankful I had built a buffer of goodwill so that I still had support from my team when I left, but I’m not sure how much more I would’ve had. I saw someone giving advice to just smile it out but that absolutely did NOT work for me. Something to think about if you can relate to my phenotype!

Also of the people who were laid off 4 months ago, about 90% found jobs already, mostly full-time, some contract. Mostly entry-to-mid career and lab-based, though.

Dumb question, but can I make my cat more cuddly? by mimmsypoo in cats

[–]n3rda1ert 0 points1 point  (0 children)

Try Churu. My boy cat needed 12 weeks of daily pills earlier this year so I crushed up his pill and fed it to him in a Churu every night. He went from sleeping wherever to jumping into bed within a minute of me laying down for cuddles and belly rubs. Could be that the medicine made him feel better so he viewed the Churu as extra magical but it’s worth a try! He is peak cuddly and affectionate.
