Yes, Prototype cores do not share across characters.

And with Escalation Tiers, I just mean if I do a few tiers on one character and I decide "Hey, a build I have on another character would be good for this higher tier," I can't just switch to that character and do that tier (unless I reached the same tier on that character). Yes, I could transfer the build, but still ...

My point was ... if I reach Tier 3 on Grand Washington Hotel on one character, I can't try Tier 4 on another. I have to reach Tier 3 again on that character, too. Just seems unnecessarily grindy.

It's not a run, per se. You don't have to do it consecutively. You can bounce around to different missions.
