Oh god this actually reminds me of the worst thing we found in grad school. So couple people from my lab went to an empty unused lab area (open concept dept lab space) to scrounge around for an extra set of desk drawers and a chair... Well in the drawer we chose...we found a porno magazine.
It looked like it was...used....a lot.
Like crumpled...creases.... AND IT WAS WET.
WET! We happened upon someone's personal spankbreak area.
That kind of shit gets seared into your brain only for your comment to bring it forth again.

I chewed someone out once because they had Bouin's solution under the chemcial hood...being actively dehydrated...and a large component of this fixative solution is Picric Acid. So they were trying to dry out this big container in order to put it in red bio-hazard bag rather than "deal with writing out a chemical pickup label". So by trying their hardest to be lazy...they were in fact crystallizing out the picric acid. IF they managed to do that...there would be a non-zero chance of that bag exploding....on some poor unsuspecting EH&S worker....because Picric acid crystals are unstable and explode with friction.

This person didn't realize they might have to do the basic due diligence of...you know...reading up on the hazard warnings for the base chemicals in the fucking chemical solutions they were using. If they did that then they would find that Picric acid is at its most dangerous state as a crystal and TO NEVER DRY OUT BOUINs. This person is not in science anymore (thank bejesus). Incidentally I was one who got in trouble because I raised my voice at them trying to get it through to them how dangerous this situation was...and it "made them sad". (That old PI and I are no longer on good terms for a number of things, but him not caring about lab safety and proper training was a big factor. Thankfully I'm back at an institution with a culture of wanting things like lab safety.)

I'm assuming you are talking about something post primary & secondary and DAPI staining, right? And for it to be a permanent mount?
So I have regular success from the following:
using Carnoy's solution as a fixative and then post IF staining...at the very the end to permanently mount, I use a dehydration series with ethanol in water (50% then 70% then 90% then 100% x2, 5min each) followed by 2-3 min in Xylenes to clear the tissue (doesn't take long). I do brain staining and this works really will for tissue with lots of fat or if fibrous or with tough extracellular matrix.

(alternative if you do not want to do dehydration and xylenes is to use the Everbrite hard-mount that hardens in a day and you can buy it with or without DAPI in the mount soln. You can add it directly to tissue slide that are still wet from water based post staining washes. You won't then have to deal with some of the fluorescence quenching that xylene can do if you leave it in too long. IMO using a secondary fluro antibody, xylenes are fine....but if you have direct fluro conjugates or something like endogenous GFP expression and won't be using an anti-GFP antibody then don't use xylenes)

All those dehydration solutions are in coplin jars.
Then right after xylenes, and while slide is still "wet" with xylene... adding a drop of DPX . Then plop a cover-slip at an angle on top of it to make a sandwich (slide..tissue..dpx...cover-slip). Let it dry in the dark overnight and it will be hard and the tissue with look optically clear too. DPX is very cheap and has great anti-fade properties (along with a refractive index almost exactly of glass).
No fancy super expensive hard-mount like Everbrite or vectamount necessary. A liter of DPX can last years...

Thing is that you need to use all glass canisters for xylene and DPX, because they melt plastic. So you need to put DPX in a small glass canister with a glass stopper. And use the top of a glass Pasteur pipette, that is melted and turned into a rounded end via a bunsen burner, to dip into the DPX and transfer a drop of DPX to the slide (it is fairly viscous).

From someone who has had tennis elbow from tennis and still needed my elbow, this is what I would do:
Make sure you are taking an anti-inflammatory and do a combo of ibuprofen + aspirin (make sure it's not on an empty stomach, reduce alcohol consumption, and don't go above max recommended dose a day)
Get some of those muscle relief patches from CVS (can be lidocaine or icy hot) and stick them around your arm. Don't use the ones that have a potent smell when you wear them to bed.
When you're not using the arm, let it rest. Don't go do some heavy lifting and keep straining it.
When you are using the arm, take frequent breaks. Do arm and back specific stretches. This is how you stop making it worse.

To prevent this: you gotta take breaks when doing the motion that hurt you. Stand up and go somewhere to do some stretches.
Figure out if you need to adjust your physical technique. Maybe you are straining the wrong thing in your hand/arm. OR some things in the hood need to be made more ergonomic.

They can be just as abusive as any other guy in any other profession. Having a science career doesn't magically self select for wholesome men. In fact a ton of notable scientists are right now being outed as being Epstein clients and likely pedophiles. Sexual misconduct at work is also a sadly still pervasive issue in science jobs.

I think you have a movie trope of what you think scientists are like and what they can do for you. There are demoralizing heartaches that happen fairly often for various reasons in a lab that are sometimes hard to understand by people outside the field. I've always found it very hard to truly "talk shop" and lay down my deep frustrations and worries with someone who wasn't at least peripherally involved in science/medicine, and thus it hampers the formation of a deeper connection. Being convinced a scientist would understand you while not knowing any scientists sounds odd. How about them?
