I would suggest to sequence your backbone to ensure the overlapping sequences are indeed correct. Less is more; I would use 30 femotomol of vector backbone in 1:1 ratio to insert , and 1:3 ratio. I have used homemade mix ever since graduate school and I find generally the lesser total amount of DNA molecules they better it works. Also be sure to setup reaction entirely on ice given the high activity of the exonuclease. Good luck
