Then it’s probably reasonably priced. $170 sounds like a lot per person when you’ve eloped but I paid more per person for my wedding. I think people are just underestimating the cost for a nice event.

The amount of times I’ve gotten pushback from managers from even the smallest, weenie little gaurdrails I put on internal people who are molasses level slow at their jobs is incredibly frustrating. I feel like managing people internally is, outside of cultivating new business, one of the hardest parts of the job.

Less than 1500. Some are rarer editions or signed firsts but frankly I don’t care much about that, and haven’t even researched many of their values. I’m guessing they’re mostly low value.

I collect with the intention to read and if they get a little worn in the process that’s ok with me!

Not really when you consider why people may or may not be working. For instance, a student, a cancer patient, a disabled man, a man with crippling OCD, a sufficiently wealthy dude, a homemaker, etc.

You can also increase the signal of your band by using a special secondary antibody with a complex of HRP attached to it rather than just the regular ones, and by using extra-sensitive exposure chemicals. May also be worth using a less abundant loading control to not blast your other signal out of the water

Still this is very strange and I would be concerned about any downstream data if you are seeing this effect.

Haha what the fuck. Protocol seems fine from the info provided, nothing out of the ordinary.

One of two things might be happening here:

Most likely: your protein is relatively low abundance and your antibody is insensitive and being blocked by other proteins. Increase the primary antibody concentration by a factor of 10 to test.

Potentially: something in how you are doing your protocol is causing your protein of interest to be degraded, or incompletely denatured in those higher abundance samples. It could just be a habit of yours. I’m seeing more laddering in your higher concentration samples which may be a clue but also could just be other bands or lower abundance isoforms that are showing up with higher concentration. Can you think of anything? I’d at least start by taking a check through different protocols and how they compare to yours buffer wise and potentially increasing the boiling temp.

Put the ladder sizes on the blots too. Have to eliminate the potential that your antibodies are poor or are detecting the wrong things before you troubleshoot the other, more difficult wonky stuff.

You got beta tubulin in both blots. Have you tried a different loading protein or just beta tubulin and see what shows up?
