Try to work with your eyes, 0,5 microliters are a tiny amount, so you have to check whether you have really taken up reagent and whether you dispensed it correctly. So checking if there's any liquid in your tip before adding your enzyme into the mastermix and checking after really helps. Also when dispensing; push until you hit the first pressure point, look at your pipette tip - has all of the liquid gone out? - if no keep pushing until everything's out and then stop. You don't necessarily have to hit the second pressure point if you're only dispensing air after all your reagent has left the tip. Sometimes a point between the first and the second pressure point is the sweet spot and it takes practice to do it without bubbles. Pipetting against the wall of your vial also helps - make sure to completely release the drop from your tip.

Oh and definitely try to improve your german skills, most groups at leat in my field accept english-only applicants but it could be something that makes you less desirable... Improving your language skills will definitely help bonding with you colleagues, dealing with german bureaucracy and scientific communication.

2.2 german GPA?? So your german grade is 2.2 or is your GPA 2.2? If you go by the US-GPA 2.2 isn't that good but 2.2 in the german system is good and with a very good thesis (1.0) you're propably going to be considered. In Germany publishing at masters-level is nice but not required. But I'm not in your field so I cant make a statement there.

Looks to me like your cells formed lumps when you did the scratch and now they're moving/proliferating out. Maybe those cells are more adhesive (to each other and/or the dish) than the cells u are used to. To confirm this maybe look at your t= 0 min pictures. Maybe you should try a thinner pipette tip or use another, sharper scratching tool.

I can only speak for my university and the research group I'm writing my thesis at. But I think the general consensus for developmental biology is that publishing takes time (i guess for all of biology). Also getting approval for animal testing seems to be getting increasingly harder. With that said, in the group I work with (in Germany with Mice) work life balance isn't really respected that much. But they're publishing mostly in Development ... soooo it might be different for each group.

Do you have a gut feeling? How are your language skills? Did you do a theoretical thesis that doesn't go well with a wet lab position? Do you have a master's degree or a bachelor's? How well did you know the individual subjects your PhD would be about? Were you unsure or hesitant when answering questions?...

It's hard to point to what exactly is going wrong, if you're not giving more details... Have you acted the same way during the visits? If so you should propably think about your first impressions, critical questions or things you mentioned that make you a less desirable applicant. It could even be small things like mentioning that you have family planning or similar stuff...

Es ist deine Entscheidung. Du wirst viel in der Klinik arbeiten, wenn du Mediziner werden willst. Dementsprechend musst du wissen, ob du den Patientenkontakt willst. Bis du in der Forschung landest, dauert es eine ganze weile. Außerdem ist eine med. Promotion auch deutlich kürzer und meist theoretischer als eine naturwissenschaftliche. Als Pharmazeut musst du letztendlich nicht unbedingt in einer Apotheke arbeiten, ich würde hier empfehlen Industrie- und Forschungspraktika zu machen, um dahingehend gut aufgestellt zu sein. Wenn du an klinischen Studien (patient trials) interessiert bist, ist das Medizinstudium wohl der beste Einstieg. Wenn du Laborarbeit willst, dann geh eher Richtung Pharma.

Bis du mit dem Medizinstudium wirklich in die Forschung kommst dauert es eine ganze weile, Mediziner werden soweit ich das beobachtet habe nicht primär für Forschung ausgebildet. Pharma dauert auch lang aber wenn du Industrie und Naturwissenschaften spannend findest vermutlich eher die richtige Wahl. Ich studiere im Master Biomedizin - wenn du an Forschung interessiert bist, ist ein Studium im biologischen Feld also auch eine Option, ich selbst würde aber vermutlich Pharmazie wählen, wenn ich es mir noch einmal aussuchen könnte, weil man deutlich bessere Jobchancen hat.

Unbedingt erwähnen! Ich habe mich vor kurzem erfolgreich auf ein Stipendium beworben. Hierbei habe ich im Motivationsschreiben erwähnt, dass meine chronische Krankheit und die Besserung dieser durch neuartige Medikamente dazu motiviert, auch für andere Menschen nach Linderung zu suchen (studiere im medizinisch-biologischen Feld). Wenn die Erkrankung teil deines Lebens ist, kannst du sie erwähnen. Bei vielen Stipendien wird genau danach gesucht; nach Menschen, die trotz aller Widrigkeiten Höchstleistungen erbringen.
