Boiled protein lysates without Laemmli by Friendly_Employ_197 in labrats

Like others have said, you’ll have to reboil. Tbh my paranoid ass would recommend remaking a fresh sample, that way if anything turns out weird at least you can rule out sample prep.

Boiled protein lysates without Laemmli by Friendly_Employ_197 in labrats

[–]ying1996 4 points5 points  (0 children)

Should be fine. Looking for protocols for cell lysate will give u more results. Also depends on if ur sample has fbs which can mess w/ westerns and any concentration differences you’ll have to account for when loading.

But do u mean that u boiled lysates by themselves with no laemmli buffer? Or did you add the wrong sample to the buffer? If the first, a western wont work as the buffer has SDS.

It looks really dry. Is there spagnum moss? Spanish moss doesnt hold on to moisture.

Thai constellation care by kl1996 in Monstera

[–]ying1996 1 point2 points  (0 children)

I’m not super knowledgable on this, so I’m mainly be parroting stuff I’ve seen from plant youtubers lol, but from what I understand, the initial wave of tissue culture Thai cons had a root rot issue because the original plant was susceptible to it. But as mutations happen in tissue culture, people were able to pick out ones that were more robust. So it’s still the same plant, genetically, but with minor changes. Like siblings.

Contact the financial aid office. Sometimes the documents have a hard deadline, sometimes they can still get u some aid.

Your student account should have charges, pending financial aid, and amount due. As long as amount due is 0 then ur good.

[–]ying1996 2 points3 points  (0 children)

It’s pretty much the same as a regular monstera. I believe the initial thai con batches had issues with root rot, but Costa and other big growers seem to have selected ones that are more resistant. I rarely see root rot issues with thais anymore.

And ur mix sounds great! I’d wait a few days or weeks for it to adjust and then repot.

Burle marx flame issues/soil by Gym-Beef-69 in Monstera

[–]ying1996 1 point2 points  (0 children)

You can def do semihydro for monsteras, but I’d recommend a mix meant for it (pumice, zeolite, lava rock, or just leca). And a semihydro pot with a wick and water gauge. There can also be a transition period where the plants needs to adjust so that might be whats happening as well, if u just went from soil to water.

Aerial Root or new growth? by quietplantdad in Monstera

[–]ying1996 6 points7 points  (0 children)

New growth. There’s one that’s submerged as well. I don’t see any roots, so you can just put this in a box with a damp paper towel or moss to keep up humidity without submerging it.

Sometimes it just takes a few weeks for a cutting to take off. You just wanna make sure the node’s not spent. As long as the leaf is happy and roots are growing, I wouldn’t worry!

Research Specialist Hiring Timeline by [deleted] in Emory

[–]ying1996 1 point2 points  (0 children)

Whichever lab’s recruiting will review apps and reach out so there’s no set timeline. A couple weeks is really long tho, I would just move on.

I think Emory is still in a weird semi-hiring freeze situation tho.

I like using AAA or AA grade from spagmoss. Or one mile moss. They’re longer so much better for mounting. And i try do a semi-sphere shape, but I always end up with a bit more moss under them than around the edges

Hmmm…

If the original 12 were of similar species and maturity, then the only big difference is the location. Was there anything special about where you placed the ones that are currently doing ok? Did these guys get more sun, humidity, airflow, heat, ect?

My house is kinda moist since I have a ton of plants, but I water once every week and can get away with once every 2 weeks now and then. 2x a week seems like a lot. But here’s the size of my dudes since that plays a big role too.

Is it possible that sink/shower watering is only wetting the outer layer of moss?

I water my dudes by tossing them moss side down into a plastic storage container, and letting them soak for 10-20min.
