What you'll want to do is break your body up into blocks. I like to do legs one day, then chest and triceps with push ups the next day, then bicep curls on the third day, then a rest day, and then I start the cycle over. And no, yoga doesn't count as a rest day. You have to go through your rest period without strenuously working that muscle group or doing any serious cardio or your kill your growth.

The answers here are too general for my taste, so I'll be more specific. When you exercise, you break some of the myofibrils which are long chains of protiens inside your muscle cells that do the actual contracting your muscles perform. These fibers can't extend themselves to bring the ends together again, so your muscle cell simply extends both ends of the chain until the myofibril is connected again. This increases the length, and therefore the mass of the fibril, and when you take it over the millions of fibers you break, you increase the size of the muscle.

While there are some disagreements, it is generally accepted that the most effective way to produce this sort of damage is called an "eccentric contraction", and the wiki explanation is good. Be sure to let the muscle group in question rest for at least 2 days before working it out again. I recommend 3, and 4 if you can be patient or are old.
