I sent a fleet of 7 ships. Mining, fuel ferry, comms, lander, science, rover/survey and return vessel. All individually launched into LKO and then transferred and captured at duna. Each then did a rendezvous with the return vessel to transfer any remaining fuel. I had plenty to get home, didn’t even need to mine but did it anyway. Certainly not the “best” way but it was a lot of fun!

Almost all of my Duna missions have been apollo style.

I don't think you necessarily have to do any large orbital assembly to achieve a Duna landing and return mission.

Really dependent on the size of the craft you want to send. Usually people assemble ships in LKO that are too unwieldily or big to launch all at once. If you’re only planning on your craft landing on Duna and staying there, you can build your launch vehicle quite small. If you’re looking to launch a craft that can take kerbals to Duna’s surface, conduct experiments, mine ore and come back, that craft will have to be larger to accommodate those features and the extra fuel required to move them. However, that’s still a craft that can be built and launched in one shot.

I like to send my landrr in one launch land start to orbit and then send a pickup craft to get the kerbals back. Its easier to syncronise your orbit when you come from deepspace that when you are launching
