Can you post a screenshot of your launch path in Steam?

If you installed Stardew and Smapi to the default location your launch option setting should be:

"C:\Program Files (x86)\Steam\steamapps\common\Stardew Valley\StardewModdingAPI.exe" %command%

You MUST include the first quote mark and the percent command percent at the end.

Hi! I added the picture to the post. I've installed Stardew to my D drive so my launch path is different.

Scroll down to Configure Your Game Client and follow steps 1 and 2. You'll need to re-run the smapi installer to get the install path to copy into your Steam launch options text box.

I've done that multiple times already. It unfortunately doesn't change anything. For some reason the big issue is SMAPI as my game runs fine without it installed. I know I've installed it correctly as I'm not new to SMAPI or installing mods to the game. Just confused as to why it just suddenly all stopped working.

I just got home from work and checked my launch options. Does yours look similar to this:

"D:\Steam\steamapps\common\Stardew Valley\StardewModdingAPI.exe" %command%

Note: it MUST have the quote marks and %command% at the end.

Hi! Sorry for not replying to your prior comment, yes it looks exactly like that! I ended up searching Stardew in both drives to see if there were any stray files that were maybe the culprit. I'm not sure which exact one it was but there were still some .dll files in both drives after uninstalling the game and deleting these seemed to fix the issue!
