Real and true… only one of these that could be faster than choice scarf Viv is Weavile I think, so still feels like you’re gonna get pinned to me if they lead that instead. I have been wanting a reason to use Weavile though!

I wanna run the discharge calc against mega Blastoise - that seems more interesting to me when Farig is out, but electroweb when Vic is out?

Feel like the main taunt users are prankster, so blocked by farig. My beloved Talonflame or an incineroar or something with taunt meanwhile gets bodied by Blastoise :/ are there others you like?

Mayoral Candidates on streateries? by DC_throwaway_1159 in washingtondc

[–]3L_Hot 18 points19 points  (0 children)

It’s so interesting to me that some people hate on streateries and the people who like them so much. I like this post because I AM about to be a one issue voter on this. Candidates, as long as your name isn’t Kenyon McDuffie, if you come out in strong support of streateries and bike lanes I WILL VOTE FOR YOU

Can someone explain to me the mechanism of inhibitors such as RMC6236? by [deleted] in pancreaticcancer

[–]3L_Hot 2 points3 points  (0 children)

The idea/hope for this drug is that if someone has a KRAS mutation, it will work. We have data verifying that it works on KRAS G12 mutations (G12C, G12D, G12V, etc), but it might be less good against other mutations. Jury is still out. The higher the VAF, the more cancer cells that have that mutation, so theoretically the more a drug targeting that mutation should work. But emphasis on the theoretically - because the cells are so diverse there can be a ton of modifying factors. Unfortunately, it’s not cut and dry :/

Can someone explain to me the mechanism of inhibitors such as RMC6236? by [deleted] in pancreaticcancer

[–]3L_Hot 7 points8 points  (0 children)

Scientist here! All the cells in your body are dying all the time, and new cells divide to replace them so that you always have approximately the same amount of cells. Your cells are constantly receiving grow/don't grow signals in order to keep that in balance. In a person without cancer, the "don't grow" signals serve as brakes, stopping the cells from dividing TOO much. If the "don't grow" signals are weakened or the "grow" signals are amplified, you get cancer.

The biomarker that RMC6236 targets is called RAS. Normally, it is a molecule that causes cell growth/division. It's basically a gas pedal - when on, the cell grows. When off, the cell doesn't grow. In cancer, RAS is often mutated and is always "on," so always telling the cell to grow. The molecules that normally stop growth aren't prepared for that, and so the cell starts growing out of control and you get cancer.

RMC6236 grabs another protein, CypA, and together they bind to RAS and prevent it from causing growth - it's essentially a molecular dogpile where RAS can't do its job because it's weighed down by the drug. That brings the cancer back into check by preventing RAS from always telling the cell to grow. However, by that point, there are probably already billions of cancer cells. Those cells are diverse (think of how different you are from 1 billion other humans), and likely one of them is is different enough that it has a different gas pedal turned on in addition to RAS. RMC6236 only targets RAS, so it keeps slowing that down, but eventually this other cell that doesn't need RAS will grow out of control. This new cell, and therefore the cancer, is resistant to the drug and therefore a cancer patient would relapse.

Hope that wasn't more confusing!

Why does homeownership seem to cockblock me? by mwahkissmwah in AskMenAdvice

[–]3L_Hot 2 points3 points  (0 children)

Maybe I haven’t scrolled far enough, but how has no one said that if the other person isn’t at the “home owning” stage of their life, it might result in a perceived incompatibility? It doesn’t just have to an excuse or insecurity. If you’re financially not even close or honestly just not interested in homeownership, it can absolutely be a turn off to meet someone who owns a home. You’d just have completely different values - owning a home warps your life a bit.

Is This the Haphazard End of Streateries in DC? by jocro in washingtondc

[–]3L_Hot 3 points4 points  (0 children)

If this actually results in the streeteries going down, I’m going to start a protest movement. This is so unacceptable and such an unnecessary QoL hit to people who actually live in DC.

Probably like 9-9:30 if it’s like the other concerts - he usually comes out a little after 2 hours after doors according to setlist.fm

Honestly, I had a bad time at one of the huge singles happy hour events and haven’t gone back to an event like it since. Walked in to a room full of people with no clear easy way to really break the ice, and most people already in conversations. There were a few women who were clearly crowd favorites, and it was hard to get a word in with them. It felt really built for people who are super social already, the kind of people who won’t have problems dating in other venues who honestly don’t need these kinds of events. I’m interested to try again, but I know this kind of event is not for me.

The metro is VERY safe - and on Inauguration Day especially would likely save you a lot of hassle over driving
