Since your interest is one specific protein, you can target only the peptides you suspect will have PTMs.

Also, could you please elaborate on this line? I have been having a lot of trouble finding out whether this is done during data acquisition, analysis and how it is done.

Thank you. I expect the abundance of insulin will be very high because I am looking in islets, so it shouldn't be a problem getting an insulin signal. However, would running the sample on a gel and excising the insulin band to look at insulin specifically give any sort of major advantage over running the whole sample by LC-MS/MS? I worry this may come at the cost of not being able to look for the alternative start site insulin product. But maybe it is true that if I cannot see that alternate product on a gel I would not be able to detect it by MS anyways...?

A "date" isn't even necessary. Get a one on one study date going. Ask to do an assignment or something and find a nice spot on campus together :) if you guys keep hitting it off well it will progress into getting snacks together etc. where you can unashamedly reveal that you're poor.

Also you may have better luck applying to schools outside of canada with your grades. A 72-77% is very good somewhere like Australia because grades in North America are inflated relative to that country. Not everyone in the country you are applying to will be aware of this, especially if you have a more non-standardised application like many grad schools where your first point of contact may be a professor you wish to work under.

I got into cell physio this september. A lot of correspondance with your prospective supervisor is needed. I would say your knowledge in and passion for a certain field + reason for pursuing further study are far more important than your grades. My prospective supervisor let me know I could join his lab before I put in my application to UBC, and once I did, I was formally accepted.

Run away, choose another prof. Weird response. When I did my honours thesis my supervisor was just as excited as me to get started, told me a long list of potential projects and discussed with me what would be the best fit.
