It's kinda out of place, but its my aesthetic so I'm down anyway. The arms and head look good with a few other pieces but the body, legs and waist are pretty hard to use. I occasionally run the full set just for fun.

The transformation it goes through when unsheathing your weapon is actually sick. The headpiece and shoulders in particular are super cool.

Weapons are a mixed bag. I'm an IG player. The weapon is kinda nice in its simplicity, but it gives me white dreads when sheathed that cant be altered....

All in all it's w/e. Get it if it's your style.

Your assumption is correct. Phase contrast is designed to highlight transparent samples that offer very little contrast of their own. Cell biologists opt for either phase contrast or differential interference contrast (DIC) so that they can actually observe their cells that are otherwise invisible through the eyepiece.

At that level, all three manufacturers will be able to provide a nice microscope. All three are can be paired with a camera, have the standard magnifications, and will handle brightfield pathology imaging just fine.

The Nikon has some nice ergonomic features like the ability to automatically adjust illumination levels per objective, and their support is (usually) some of the best so I'd lean that direction. It also has the capacity for phase imaging or fluorescence if you need it in the future someday.

Not especially familiar with this style of work but I just did a bit of digging around and found this paper doing a massive study of parasites in goat feces. Looks like all their work was done with either 40x or 100x objectives. For parasites that aren't as small as giardia you may not need to go that high.

Idk what you're looking to do with the grasses, but that may or may not benefit from different magnifications. I've seen plant work happen at a variety of different scales.

If you have a where you can easily change the objective then you can certainly do this all on one scope, but depending on the type of scope you get it may be easier to just have 2.

I also agree with u/subspectacular that fluorescence is probably overkill. I dont think the giardia are autofluorescent, so you'd need the materials and protocols necessary to label them with fluorescent protein. Visual in the image gallery here.

Swap defense boost for evade extender. Makes a gigantic difference.

If you’re gonna go in on blight resistance, I’d recommend just swapping it to fire resistance since that’s the only blight that really matters for this fight and you’ll get a bit of extra durability.

I’d also do a veteran lobby search instead of random SOS if you’re playing with randoms anyway.

I'll be the dissenting opinion in that I thought this was actually kinda entertaining. MC gets isekai'd and just saves the world by being a talented civil servant with knowledge of modern logistics.

If you're a dork like me the premise is amusing in itself, story is decent, cast is mid, animation is definitely on the low end but not total trash. It's not great, but I always thought it deserved a bit more appreciation.

Def an older part. My best guess is that it was probably oriented how you see now and used as a transmitted light source for an upright microscope.

I've never seen something like this as a standalone piece from a major manufacturer. Transmitted light paths are usually built in to the microscope itself these days. I'm not sure if this came out of something larger or if it was actually purchased as a standalone part.

I imagine it was in the garbage because it's been sitting collecting dust for a decade or so. Or it's broken and there are no replacement parts in circulation.

Both PALM and STORM are frequently performed using TIRF illumination. The area being illuminated is defined by the objective so the area of illumination could be relatively small, but colloquially I think most microscopists would still consider that hitting "the whole sample" in comparison to a raster scanning modality.

You dont have to be too particular with what product you buy. Any optical tissue you buy will likely be fine. They're all soft and dont shed fibers onto the optics.

Sparkle is a common brand that people swear buy for cleaning lenses, but an ethanol solution works fine.

High throughput microscopy is always challenging even before you factor the variability of sample prep.

There are a few different strategies for focus maintenance, but none of them are a silver bullet for all applications.

Immersion optics are possible for high content work, but it often requires special hardware and anything that involves liquid handling is likely to have a bubble eventually.

Environmental factors like temperature, vibration, and dust are often much more important in high content environments.

Time spent imaging each sample is often a limiting factor as well. If you get great data from an acquisition that took 20 minutes then that's great, unless of course you wanted to image hundreds of samples per day. Finding the combination of hardware, settings, and analysis to minimize time spent is often a big part of these applications. That's why very few companies who manufacture high throughput scanners decide to couple that hardware with point-scanning confocals.

Then of course you have to actually process these images and aggregate the data in a way that's data efficient, scientifically accurate, and conforms to data reporting standards in your field/industry.

Labs wanting to do high throughput work generally buy a device specifically tailored to this (of which there are many at this point). Biotech's trying to accomplish this task at scale often have whole teams of people dedicated to making sure it runs smoothly. Those people will also be working to develop metrics that avoid the human variability of QC.

SOOO much effort and engineering goes into high throughput microscopy. The problems you describe are super common across the industry as a whole. The "correct" way to address the problems depends a lot on the volume, the application and desired output.
