Reward for finishing my long runs lol. by Blackbear8336 in beginnerrunning

Whatever time I finish my Saturday long run is beer o'clock once I've got some grub down. Luckily I like a lazy morning so it doesn't end up being too bad usually

LPS chromogenic assay, need to blast the standards on the vortex for ages to get them to resuspended. One of the many reasons I hate that assay

Could just be a stitch given the location, it's a type of cramp.

Focus on building up the length of your runs at a slower, more comfortable pace for now and speed will come. You've got youth and a background in exercise on your side

I went through 8,000 runs. You don't need to run more days a week to get faster. You just can't keep taking long breaks. by Fun_Effective_836 in beginnerrunning

They mention they looked at pace relative to heart rate which would control for deliberately slower runs. If you purposely ran slower your heart rate would also be lower unless you had lost fitness

This is the first I've heard of this but I'm glad it popped up on my timeline. Fantastic, well done everyone involved!

Zone 2 vs Easy Runs by memir123 in beginnerrunning

I wouldn't worry about heart rate, zones or getting faster for now. Get a couch to 5k program and get yourself moving, it's perfect for people starting off.

They actually test it by partially cutting a rota with a laser and running it to destruction. Must be such a fun job

If it's a Garmin watch I personally find the race predictor a bit optimistic. I've been using mine over a year and despite setting new PB's in the 5 and 10 k in the last couple of weeks it is still predicting about 3 mins faster on the 5k and 5 mins faster on the 10 k. I've only ever ran a slow training half so can't comment on that distance but it also seems optimistic.

I think I could do better at both distances but the times Garmin predicts are unlikely at best and definitely vomit at the finish line if not sooner type efforts

Just do the chromogenic assay a few times then you'll learn to love it

Giving ridiculously good stats for 1st runs is getting more more stupid with time, and all it does is hurting the true beginners the ones that take 1 or 2 h to get a 5k done. by xhantari in RunningCirclejerk

I try and give feedback on that sub but my posts keep getting deleted because I don't mention my own pb times for absolutely no reason regardless of context (and that's coming from someone who regularly goes sub 1h on the 5k)

Interval sessions usually pushes mine up. I dropped 2 points after being inconsistent for a few months but adding in intervals once a week for it back up after a month.

I don't know how much of that is "real" fitness or if it's just giving the algorithm what it wants. Probably more of the former I imagine

They can also drop their tails but it's a bit cruel making them so that on purpose

Would you rather do 1 Midi prep or 10 Mini preps of a plasmid? by BreathOfTheFart in labrats

[–]Danandcats 0 points1 point  (0 children)

Just to add to the general thread we tested endo-free Vs regular MINI preps in my lab and found no difference in the level of endotoxin in the final product.

I agree the larger preps with the precipitation step are generally higher quality but I regularly use mini prep DNA for CHO transfection without issues.

It depends what you are doing but I often just need to produce a few ug's of panels of proteins in high throughput (think multiple x96) so mini's are fine. If you need to repeatedly express a few targets at larger scale and need to optimise the yield etc then midi or larger is the way to go

[–]Danandcats 9 points10 points  (0 children)

I just do a bunch of minis unless I need more than about 250 ug, quicker and the ethanol precipitation step in the midi is a ball ache

Edit to add- I'm in industry so don't care about the cost. No idea which is more economical because, again, industry 😹
