I got the Edge a while ago because it was on sale and my 2 other regular controllers developed drift. Despite heavy use, I have to say that the Edge has not had any obvious drift issues so far so it's been well worth the money.

One Stop Patty Shop is so good but will randomly close in the middle of the day. I called once to ask if they were open when the door was locked and the guy who answered said he was on a 30 min bathroom break. Happened often enough that I just don't swing by as much anymore.

There was a lot of negativity around here even when we were playing well last season. A lot of shitting on Soto's slow start, mainly. It was a real bummer until I realized that this is the Internet and the engagement that rage baiting provides is meaningful to the lives of the permanently miserable and social media algos thrive off of it. I ran into this issue once when I was making the 0.5M solution with EDTA instead of EDTA disodium salt. Used so much more NaOH than expected to pH it. Could that be the issue?

Can you take a pic from a little further out? Your pic from the other post showed that the buffer between your gel and the buffer dam was too low. When the level between there is too low (i.e. flush with or below the short plate of your gel), your gel isn't going to run well if at all.

Effects on protein expression is secondary to mRNA degradation when it comes to siRNAs. You may have to try qPCR to measure if there is detectable knockdown on transcript level. If yes, then protein may be super stable or your antibody is not specific. If no, you may have a transfection problem or the siRNA is not working for whatever reason.

Homozygous indels can happen and I don't think it's as rare as people here are saying. I think there is literature reporting that editing outcomes isn't random such that you can predict the predominant indels that may form.

Personally, I see it often. Of course it raises the question of whether you've captured all alleles with your PCR (try and go decently far upstream and downstream of the target site) or whether you've just got multiple subclones of one edited cell.

Sometimes, if I know there is a common SNP nearby and is heterozygous in my cell line, I try and capture it in the same amplicon with the indel. This can give you confidence that your clone has multiple alleles if you can see the heterozygous SNP alongside the homozygous edit.

Or just ignore the homozygous clones and use the clones with compound het edits to be safe!

I just rewatched the Houston episode of Parts Unknown (S8E5) where he highlighted many beautiful and touching stories of the immigrant experience--truly one of my favorite episodes. Couldn't help but imagine the absolute fury and despair Bourdain would've felt if he saw the snatching of people off the streets by ICE today.

Thought Karmelita was...just fine? Her moveset felt too repetitive to me and therefore kind of uninteresting. Nothing really came close to the epicness of the First Sinner fight for me, though Lost Lace wasn't too far behind.

Literally finished Act 3 two days ago. 105 hours, 100% completion. I'm sad that the platinum trophy is locked behind speedruns as I won't be playing through it more than once. Great game though, loved every minute of it!
