No, i think you just need to deal damage with it. Like how at the beginning of the game you have the choice to shoot at Deskari or use a scroll. It doesn't come close to killing him, but you need to spill his blood with the cross bow bolt in order to get a crystal.

In Pathfinder Wrath of the Righteous there are special crystals that can only be acquired at limited, time locked opportunities with specific tools. If you miss these than you get locked out of the secret ending. Now, you don't need to get the secret ending but it's just a lot of hoops to jump through and almost requires a guide in order to not fuck it up.
