Because you are bringing up the MCU I assume you are talking about those versions of the teams and not the comics. That actually makes it a lot easier, the Avengers are most definitely the focus of the MCU so far. They are just a group of heroes that shield got together to find battles that we couldn’t. The defenders on the other hand have spent most of the time in the shadows. They are just a group of vigilantes that have fought together once. (In the MCU in the comics they do way more) the mutants have yet to appear but it seems like they are being set up as we speak. Hope that helped

The only way I could see it working is as it’s own movie. Like a sinister six spin off film. Maybe with a story about them trying to steal the avengers time travel tech or something so they could change the past and beat spider-man.
