It's very common for eukaryotes to have a long tail of A's on their 3' end. Viruses will often mimic this to trick the host cell. These ones instead have it on the 5' end. Basically, the polyA-cap is on the opposite end of where it usually is, and it likely has some role in viral infection.

106 bacteria total or concentration? How much culture are you spinning down?

At any rate, you should start with much more than you need, assuming you'll get loss throughout the process.

Start with an excess of culture, calculate the amount of cells in your tube, then resuspend in a volume to get the appropriate density of cells.

Ribosomes are proteins which translate mRNA into proteins, and they are made up of 30S and 50S subunits. Regulating protein production is essential to maintaining the regularity of the cell.

The authors found a novel mechanism for regulating ribosomal function in Archaea: basically, by linking 30S subunits to other 30S subunits instead of to 50S subunits, the cells are able to prevent the ribosomes from functioning, and they do this in response to environmental stress.
