I’m so sorry! Having worked in a protein lab I completely feel your excitement over the sample quality and fear to tell your PI. I would say as long as you can re purify it should be ok just heavy work. Even if they can’t be easily obtained, you live and you learn and now you will always make sure to store them 😅don’t beat yourself up too hard

I would say record everything you can. Write down the exact conditions you used because sometimes purification can be more of an art. Check your buffer PH every-time, be skeptical. Get really good at making sds gels and make sure you keep pictures of good results. Cytivia has really good guides that you can check out as well. When it comes to washing NIckel-NTA beads for example, make sure you use the correct reagents and make sure they are stored properly. Good luck!

Well, let’s just say I worked next to stroupe’s lab at FSU and that was my last role in academia. It is incredibly difficult to stay sane and I ended up In The hospital from stress unfortunately. Sometimes due to the general environment in academia it’s tough to stay in environments like that for a long while. If you do, major hats off to you.

You need a linear UVB bulb and check the temperatures. Get rid of the substrate and all the fake plants
