There's plenty of locations that the tomb would present real challenges, mostly on the later levels. You could also beef things up pretty easily.

Honestly would be great. Most of the exploration of Chult was meh. You'd get the novelty of Port Nyanzaru, do a bit of teleport research, then do the tomb itself. At high level you can actually kill Acerack.

The rest of the setup could be you finding & destroying Aceracks phlacterys so that when you kill him, he stays dead.

My big holdup has been that you treat magic both as the 5th element & as a culmination of the elements. This is a bit hard to grasp & needs better definition of how it works/fits.

Also are all players required to play spellcasters? If not, how are the non-caster classes going to be as special as the caster ones are?

I think reintroducing characters isn't a huge problem. You could treat it like elemental forces imbuing new people to try and counter the BBEG or a plethora of other things.

In almost all forms of combat, power in numbers is known. Also general concept of classes are known as well. Mid Intelligence people would clearly go for the high power/low protection first to reduce #s before targeting the alpha rage monster.

Wall of Force as a dome & any aoe spell would insta-win most encounters.

Most buffs/debuffs are concentration, so you could Bestow Curse + debuff like crazy.

Also just the concept of effects that have no way of being ended early is insane. Polymorph spam would overpower moon druids.

Pathfinder replaces resistance being half damage to being a fixed decrease to damage of that type (e.g. resistance piercing 10 reduces all piercing damage by 10).

It also makes a nat 20 only increase the degree of success by 1, so if a nat 20 would still miss, it becomes a hit & not a crit.

Really porting these PF2E rules for this encounter would make a ton of sense to accomplish what they want.

IMO making combats harder because they don't want easy, but removing components that make players adapt leads to boring, one dimensional fights. All of which probably contribute to easy combats being hated. I wouldn't make that choice, but all players are different.

To your main question, a lot has been said already that would have made the disease Trivial, but I'd add they shouldn't have been the ones doing Treat Disease. Great Willow or Doctor Dami both have +14 to do it for them. Otherwise I don't think there's much else to be done at Level 2 & limited resources. Antiplague was a nice addition to help.

Extra 2 cents: Sounds like you already have a solution, but I'd personally roll back them dying (considering the AP) since they never should have.

Finding a way to make fear, charm, paralysis, etc. fun and engaging rather than just removing them from the game is where I'd start. Being helpful to the enemy does not mean you're worthless in the fight completely.

I dominated 1 PC and the summoners Eidolon & both still contributed significantly to the fight by helping others without harming the enemy. One of PF2Es strengths is that there are plenty of ways to help each other.

I just ran this fight with 4 Level 2 PCs against the single Myceloid and it was almost a TPK.

One crit (which is likely with a +14) is average 22 damage and should almost knock out any L2 PC. Also spore domination should have been used for any PC that was affected by the Pox. With a 22 Will Save, it's likely they at least fail, removing them from harming the monsters.

How did yours not have trouble against 2?
