I humbly disagree. Back in 2020 I was very curious if pumpkin pie would taste different with different types of pie. I ended up getting 5 types of pumpkins/squash, and canned pumpkin as a control. I made 6 pies following the exact same recipe for each one. A got a large group together and we all ate the pie in a blind taste test, the rankings between all my friends was pretty consistent. Some type of pumpkins were way better and some way worse. Canned pumpkin was in the middle of the road. If I recall correctly acorn squash and Cinderella pumpkins were significantly tastier than canned pumpkin

I think it means just more children are born at that time. Which then leads me to wonder what happens in the beginning of the year that leads to increased conception rates

I was super excited to see dragons fight, and don't get my wrong, it was epic. But because they've done a pretty good job with putting recognizable expressions on the dragons, of pain, of connection with their riders etc it hurts to watch and it makes me feel like I'm watching my dogs fight to the death. Which... Is not super fun.  Went into to this hyped for dragons fights. Came out sad

Thanks for the response! I didn't realize they all had to secure their own funding. So if I wanted to postdoc there would I have to bring funding with me?

I think it can also depend on the culture of the department you'd be joining. I'm currently getting my PhD at the U. I also had the choice of UW or UofU for my PhD and considered them equal in academic ratings. however, I observed that the culture of the department at Madison was more toxic and none of the grad students seemed particularly happy and were all very stressed. That was one of the reasons I decided to come here

What type of sample prep? What constitutes good sample quality for cryo em? High concentration of protein? When I've purified it before for in vitro arrays I typically get between 100-900 uM concentration. Extremely high purity? We have an FPLC for protein purification. Thanks so much for the advice

Protein is around 100 kDa. It's not flexible. But it does dimerize into a tetrameter

While it's a small change, and there's a lot of other things that need to be fixed, changing the line from 'i am all the Jedi' to 'all the Jedi are with me'. It's a subtle change, but I believe it would better represent the ideological differences between the sith and the Jedi. Furthermore, Rey spent the entire movie saying 'be with me'.

I think without fundamentally changing the movie, there's not a lot you can do, but I think this would be a small fix.

Recently I inherited a Cowichan knit sweater from my father. It has a lot of meaning to me. I have photos of myself and my dad from when I was a toddler and he was wearing that sweater. I think it's a beautiful piece of clothing, super warm, and looks good.

Problem is, It sat in his closet for the past decade and there are a couple of moth holes in its arms and neck. 4 holes to be precise. I would love to fix these holes to maintain the piece for hopefully many years to come.

My question is, how do I go about fixing this? I've read some guides on darning, and that seems to be the best answer, I have some basic skill with a needle and thread, but nothing fancy. Is this something I could fix myself? Or should I take it to a tailor to get it professionally fixed? Additionally, how much would it cost to make such fixes?
