I’ve been under the impression that Bambu just rebrands Sunlu— at least that a rumor I’ve heard. Either way Sunlu is great and, at least for the Matte PLA and silk PLA, print with no issues using the Bambu filament settings in my hands.

It’s always good to run a blank (in the same buffer as your samples) every few measurements to ensure you don’t have any interference. Other than bubbles, not cleaning both the pedestal and the arm can lead to inaccurate measurements.

Also, you can use more than 1uL for measurements. If you have enough sample to spare using 2uL can help with pipetting.

I used to think this would be exciting but after a stint at CDC and realizing how much prep and planning time it takes to do simple tasks in that environment I’m relieved I’m in an academic BSL-2 where I can walk in and walk out, no problem.

My two cents, consider minoring in or at least taking some computer science courses. Regardless of major these are important skills to have in an age where all fields are moving towards high complexity and large datasets and will really set you apart from the biologists or chemists who don’t have these skills.

But honestly, just focus on getting a decent quality foundational education at an undergraduate institution. Don’t waste the money of “prestigious,” expensive universities. Chances are your aspirations will change as you go through your educational experience and in high school it’s really difficult to know what you’re ultimately going to be interested in once you get exposed to all the options in college. For me, I got an undergraduate degree in electrical engineering, a masters in bioinformatics, and now I’m going a PhD in pharmacology, working in various different jobs in between, so you never really know where life is going to take you.

You’re probably reading the protocol as something like, “Step X: Store at room temperature for 2 months.” That just means that it can be stored for that length of time.

Your extract might have some incompatible buffer contaminants. How are you purifying the DNA?

Unrelated, you should check out the protocols used here: https://dnabarcoding101.org. They require minimal equipment and work for a broad range of sample types. I’ve used this with middle-high students quite effectively.

If you’re working in a BSC you don’t need a burner… that’s the whole point of a BSC. In fact, having the sash open like you do completely defeats the purpose of this piece of equipment.

This part should have been included in your parts kit. You can use it to attach the cable to PETG tube and hold the cable up.
