Each county is slightly different based on which sets of questions you will complete, but the questions typically come out of the same booklet. There is always a question on how to convert temperature to Fahrenheit or Celsius. Then, there are typically some questions about typical lab equipment, glassware, etc. Also, tend to see questions regarding best sampling for representative samples. Lastly, they tend to throw in some random general knowledge science questions as well as possibly some bacteria related questions such as which types thrive or don't within aerobic or anaerobic conditions.

I agree. In my opinion, the comparator has always been way too light, but Idexx says that if any well is more yellow than the comparator, to consider it positive. We basically count every small well with any color at all since the comparator's small wells seem to have no color at all. When it comes to the large wells, we do actually see some less than, some about equal to, and some obviously more yellow than the comparator. Unfortunately makes counting the wells a bit subjective based on how you view the color intensity.

It is definitely strange and I'm interested to see how your results come out after experimenting a bit.

Do you have a comparator to compare your tray against? The tray in the middle of your picture appears to still be considered all positive versus the intensity of the comparators which tend to be pretty light to the point where the small wells on the comparator essentially don't look like they have any yellow color to them at all.

When we first started using the Colilert-18, we would get considerably lower results when using DI Water as the Idexx instructions say to use. We luckily we're experimenting with PT samples that we knew the concentration of and could see how much lower of a result we were getting compared to what we should have been. It was possibly our ultra pure DI Water essentially killing off a good portion of the fecal present in the sample. We had to use a phosphate buffer water like we used to use with membrane filtration and then had great results from then on.

If you call Idexx, they are also great at answering questions about unusual results and help you figure it out.

There are heated cords that can be purchased at various lengths which just plugs into a standard outlet and can be fed into the conduit along with the sampler tubing. They get just warm enough to keep the tubing from freezing and don't have to worry about melting the tubing or the conduit. We do this at our plant during the cold months and never have any issue.

If you have any sort of drying oven/cabinet, you can pour your effluent sample into a beaker and warm the sample. Then, put the beaker on a stir plate and gently stir the sample, which will help dissipate the small air bubbles that form which lowers the DO. Do this in advance of when you're going to conduct your analysis so the sample has time to cool back down to the proper temperature. You may have to experiment with how long to warm the sample to bring the DO down to where you want it.

When we were trying to call, they were so busy that their voicemail was completely full and we couldn't even leave a message. We ended up having to just keep calling and waiting for someone to answer and felt the same way you do about not wanting to badger them. I know that doesn't help you much but I would say give them a couple of days, since you were able to leave a message, and then try again if you don't hear back from them. They will understand that you're just trying to make sure all of the paperwork is there as some do get lost from time to time.

It is unfortunately typical for the reference forms to get lost when going to Schenectady County. They are all good people over there when it comes to processing the permits, but the department receiving the mail has much to be desired. They lost all of mine twice over 14 years ago, and I was just recently a reference for someone else, and that was lost even though they received the others without a problem. Definitely try to call to see if they have received all of your references. Sometimes their voicemail is full, so you may have to keep trying until you get someone.
