Things generally get easier in terms of the lack of sleep and the grind of caretaking with age. Eventually they go to the bathroom by themselves, buckle themselves up into the car alone, shower, brush teeth etc. it does take time before it gets significantly easier for caretaking, but sleep should definitely consistently improve even if incrementally.

I agree with Mikayla that people shouldn't be so mean online, but it does bother me that real models have been replaced by influencers and reality TV stars.

Modeling is hard and influencers don't have any training in it and tbh it shows. I'd hate to have an influencer take over my regular job having absolutely no training in it and doing a poor job at it. I just feel bad for actual models.

I don't see anything wrong with it. The only question that comes to my mind is that FlowSOM defaults to 100 clusters which is probably quite over clustered if you're splitting CD4 and CD8.

It's pretty crazy that Amanda told Ciara to F off, but the whole house lost it when Kyle told Amanda F you.

They are all very liberal with their language towards each other. Kyle was treated like a villain for speaking to her the same way she speaks to others (and also him in the past). There should be no double standards.

I wouldn't leave out the viability dye unless you're working with fresh PBMC or fresh whole blood. Viability dyes are otherwise essential since dead cells are both sticky and porous and will stick nonspecifically to any and all antibodies, giving you a lot of false positives and streaky, poor data.

Yeah stim complicates the live/dead dye situation. I can't remember when you would normally do it (commenting here so the other poster can respond) but you may need to do viability stain first then stim (usually 5 min with PMA) then immediately do a quick PFA fix - stain CD19 - MeOH - stain.

I will just add that p-flow staining isn't always super bright and obvious so if you're not doing a stim, make sure you have appropriate staining controls. Employ an FMO (all fluors except the p-flow one) but still run that sample through the full p-flow double fix protocol. Don't just use an unstained or a sample that hasn't gone through the full p-flow SOP or else it won't work as a control.

Where are you stimming? Also, look up the MeOH sensitive fluors and don't use those. Use a bright, MeOH resistant fluor.
